两亲分子聚集体调控的荧光传感器——选择性传感或区分识别

摘要
两亲性分子聚集体在水溶液中可提供疏水微区,增溶脂溶性分子,在药物缓释、生物成像、化学生物传感等领域有着广泛的应用。本研究小组利用两亲分子聚集体增溶结构特异的荧光探针分子,制备了一系列超分子荧光传感器。研究发现,两亲分子聚集体的聚集态变化可以很好的调控所增溶荧光探针分子的荧光发射。外来分析物的引入,通过与两亲分子聚集体的静电作用、疏水相互作用等引起聚集体聚集态的改变,进而引起所增溶探针的荧光发射信号的改变,从而实现对分析物的选择性检测。当所选用的探针呈现多发射峰时,外来分析物与聚集体的非共价相互作用可引起探针的不同发射峰的不同模式的响应,进而呈现多波长交互响应,从而实现对相似分析物的指纹图谱识别,并利用PCA等数据分析方法实现区分识别。
Amphiphilic aggregates could provide hydrophobic core for encapsulating lipophilic molecules,which has been widely used in drug delivery,bio-imaging,and chemo/biosensing.Our group has used this strategy to encapsulate fluorophores with specific structure to build a series of fluorescent sensors.The amphiphilic aggregates have proven to be able to modulate the photophysical properties of the encapsulated fluorophores by varying aggregation states.When the analyte interacts with amphiliphic aggregates via non-covalent interaction such as electrostatic interaction or hydrophibic interaction,it will induce photophysical property variation and realize the sensing of the added analyte.If the encapsulated fluorophore emits multiple peaks,the analyte-induced aggregation change could lead to fluorescence variation at different wavelengths,resulting multiple wavelength cross-reactive responses,which can be further used for pattern recognition to discriminate similar compounds.
