麒麟菜寡糖的制备及其抗过敏活性研究

摘要
麒麟菜为红翎菜科植物,富含胶质,可提取卡拉胶。本文以假交替单胞菌属pseudoalteromonas sp.ASY5为菌种,经发酵得到可降解卡拉胶的粗酶。麒麟菜粉末溶解在蒸馏水中,直接加入卡拉胶粗酶进行降解;降解后的麒麟菜样品经离心取上清浓缩、乙醇沉淀、真空冷冻干燥得到寡糖。寡糖复溶于纯水,制得0.3mg/mL麒麟菜寡糖,然后分别稀释成不同浓度。以DNP-BSA为过敏原刺激RBL-2H3细胞,建立过敏反应的细胞模型;细胞活力测定实验结果显示,麒麟菜寡糖(浓度25~100μg/mL)对RBL-2H3细胞活力无明显影响。细胞实验结果表明,DNP-BSA刺激敏化的RBL-2H3细胞经麒麟菜寡糖处理后,脱颗粒和组胺释放的抑制率分别为34.2%和45.9%;麒麟菜寡糖能显著降低细胞因子IL-4和TNF-α的表达,且随着寡糖溶液浓度的增加,抑制效果越来越明显。此外,麒麟菜寡糖能够显著减低原肌球蛋白致敏小鼠血清中免疫球蛋白IgE和IgG1的水平。
Eucheuma,rich in pectin,is a kind plant of family solieriaceae,which could be used toextracted Carrageenan.In the present study,pseudoalteromonas sp.ASY5 was used as bacteria,carrageenan crude enzyme solution was obtained by fermenting.Freeze-dried Eucheuma powder was suspended in DW,and then mixed with the carrageenan crude enzyme solution.After the mixture was hydrolyse,then centrifuge it.The supernatant was concentrated and precipitate with ethanol.The crude oligosaccharide was obtained after the freeze drying of supernatant.Eucheuma oligosaccharide was dissolved in DW at0.3mg/mL then respectively diluted into different concentration.Take RBL-2H3 cells as allergy model and allergen was DNPBSA.Cell viability assay shows that oligosaccharide concentration 25 ~ 100 u,g/mL has slight significant damage on the vitality of RBL-2H3 cells.Experimental of mast cells show that Eucheuma oligosaccharideinhibited B-hexosaminidase,histamine by34.2%and 45.9%,respectively.Eucheuma oligosaccharide also suppressed the expression of interleukin-4(IL-4) and tumor necrosis factor-α(TNF-α) in antigen-sensitized RBL-2H3 cells.Furthermore with the increase of the oligosaccharide concentration,the inhibitory effect more obviously.In addition,Eucheuma oligosaccharide could obviously decrease levels of IgE,IgG1 in serum from TM-sensitized mice.
