‘美人'梅不同扦插时间嫩枝扦插叶片内相关生理生化变化

为研究不同扦插时间‘美人'梅扦插后插穗叶片的相关生理生化变化,以‘美人'梅半木质化嫩枝为材料,分别在5月份和8月份采用2500 mg·L~(-1)IBA处理插穗,测定扦插后不同时期叶片内可溶性糖、可溶性蛋白、淀粉含量和过氧化物酶(POD)、多酚氧化酶(PPO)、吲哚乙酸氧化酶(IAAO)活性的变化。结果表明:①母株叶片内的IAAO活性较高可能是导致‘美人'梅生根率季节性差异的原因。②生根过程中,可溶性糖含量均呈现"升高-下降-升高"的变化趋势,而淀粉含量均呈现先下降后升高的现象;8月份可溶性糖及淀粉含量峰值点相比于5月份均存在滞后现象,可溶性蛋白含量变化趋势相比与5月份不明显。PPO活性在5月份与8月份呈现相反的规律;8月份,POD活性变化趋势不明显,且其活性较低,IAAO活性较高,可能是导致其生根率较低的原因。研究表明:母株叶片内的IAAO活性以及扦插生根过程中,叶片内的3种营养物质含量和3种氧化酶的活性及其相关变化趋势与扦插生根密切相关。5月份,母株叶片内的IAAO活性较低,生根过程中叶片内的可溶性糖及淀粉含量峰值提前,POD活性出现两个明显的峰值,LAAO活性较低,可能对生根有利。
The objective was to study the related physiological and biochemical changes in blade during rooting of Prunusmume‘Meiren' in different cutting date.Contents of soluble sugar,starch,soluble protein and activities of POD,PPO,IAAOin phloem at various rooting phase were measuredin asimplerandom replicationexperiment.The semi-hard wood stems were collected and treated with IBA at a concentration of 2500 mg·L~(-1)with water dipping as control in May and August respectively.The result showed:①The high IAAO activity in matemalmay cause cutting date effect,②uring the rooting process,the soluble sugar content increased at first and then decreased,and it increased again at last,while the starch content fall before increased.The peak of the content of soluble sugar and starch during the cutting in August compared with that in May existed hysteresis and the changing trend of soluble content was not obvious.During the rooting process,the activity of PPO changed on the contrary in May and August,at the same time the POD activity was lower and IAAO activity was higher,and the variation trend of PPO activity was unconspicuous,that might be why the rooting percentage in August was so low.Our results suggested that the IAAO activity in maternal and the threenutrients and three oxidases during cutting were closely related to rooting.In May,the IAAO activity in maternal was lower and in rooting process the content peaks of soluble sugar and starch were in advance and POD activity appeared two obvious peaks while IAAO activity was lower,which may be favored for rooting.
