5/6肾切除大鼠慢性肾功能衰竭经典模型的建立

摘要
目的:通过动物实验研究探讨寻找建立接近临床实际,适用广泛、统一、简便、稳定的CRF动物模型,对研究CRF的发病机理,探讨组织形态的变化与生化指标及临床表现的相互关系,为深入到细胞和分子水平研究CRF,筛选有效药物,阐明疗效机制,提供符合人类自然发病病理表现经典动物模型。方法:采用5/6肾切除建立CRF大鼠模型,分为模型组、未手术组(空白组)、假手术组(正常对照组)。观察各组大鼠血清尿素氮(BUN)、肌酐(Scr)、肾组织HE、masson染色病理切片观察。结果:5/6肾切除大鼠血清BUN(P<0.01)、Scr(P<0.01);病理切片残余肾脏组织纤维化存在。结论:5/6肾切除大鼠慢性肾衰竭BUN(P<0.01)、Scr(P<0.01),病理切片示残肾纤维化存在,符合人类慢性肾衰竭肾脏的病理学特征,证明造模成功。
Objective:through the animal experiment research for establishing close to clinical practice,and is used widely,unified,simple and stable animal classic model of CRF.Methods:5/6 nephrectomy is adopted to establish the CRF rats model,were divided into model group,positive control group,at the same time a control group(normal control group).Observe each group rats serum urea nitrogen(BUN),creatinine(Scr),kidney tissues HE and masson staining pathological observation.Results:5/6 nephrectomy in rats serum BUN(P < 0.01),Scr(P < 0.01);Residual kidney pathological tissue fibrosis.Conclusion:5/6 nephrectomy in rats with chronic renal failure BUN(P < 0.01),the Scr(P < 0.01),pathological section residual renal fibrosis,proved that building success.
