低温压力下斑马鱼受精卵细胞凋亡相关基因的表达分析

环境温度影响鱼类的生存,这种影响在受精卵卵裂阶段更加显著,发育温度过低常常导致受精卵死亡。然而,低温压力导致受精卵死亡的机制尚不清楚。本研究中,将斑马鱼受精卵分别放置于常温(28℃)和临界致死温度(20℃)中,发现在临界致死温度(20℃)下斑马鱼受精卵发生一次卵裂的周期明显增加,并在后续的发育过程中出现停滞。利用RT-qPCR的方法,对常温(28℃)与致死温度(20℃)下500细胞期与1k细胞期的受精卵中参与细胞凋亡的相关基因c-jun、gadd45αa、p21,p53的表达水平进行检测。结果显示,低温下c-jun、gadd45αa、p21的表达量显著上调,相对于常温下表达量升高30~60倍。但是p53的表达量并未发生显著的变化。该研究结果表明,斑马鱼耐受低温的极限受一种不依赖于p53基因表达量水平的细胞凋亡现象的影响。
Environmental temperature affect the survival of fish, this effect is more pronounced in the cleavage stage, low temperature often led to the death of the oosperm. However, mechanism of the death under a cold stress is unclear. In our research, Zebrafish embryos are incubated under normal temperature(28 ℃) and critical lethal temperature(20 ℃) respectively, low temperature results in long cleavage cycle and embryonic arrest. Using RT-qPCR to detect the expression of apoptosis-related genes: c-jun, gadd45αa, p21, p53 in the 500 cell stage and 1k cell stage embryo placed in normal temperature(28 ℃) and critical lethal temperature(20 ℃) respectively. Our data showed that the expression of c-jun, gadd45αa, p21 was upregulated in cold environment. But the expression of p53 showed no significant change, indicating that the limits of cold tolerance of zebrafish is effected by an apoptosis phenomenon which does not depend on the p53 gene expression level.
