人参提取物对于α-MSH作用的小鼠黑色素瘤B16细胞黑色素生成作用机制研究

摘要
目的:探讨人参提取物抑制细胞内黑色素生成作用机制。方法:采用碱裂解、酶促反应、免疫印迹、流式细胞术等方法检测人参提取物对于小鼠黑色素瘤B16细胞内黑色素生成以及氧化应激的影响。结果:人参提取物能够降低α-MSH刺激的B16细胞内黑色素含量,抑制酪氨酸酶活性及其蛋白表达;降低活性氧含量,提高SOD酶活性和GSH含量。上述作用与ROS抑制剂NAC相类似。结论:人参提取物可能是通过抑制氧化应激而抑制黑色素合成发挥美白作用。
Objective:The purpose of this study was to investigate the molecular mechanisms of ginseng extract on melanogenesis in B16 mouse melanoma cells.Methods:The effects on melanogenesis and oxidative stress of ginseng extract in a-melanocyte-stimulating hormone(α-MSH)-treated B16 cells were determined by alkaline lysis method,enzymatic reaction,western-blotting,and flow cytometry assay.Results:Ginseng extract significantly decreased melanin content,suppressed cellular tyrosinase activity,and reduced expression of tyrosinase protein to inhibit B16 cells melanogenesis induced by(α-MSH).They also depleted cellular reactive oxygen species(ROS) generation,increased superoxide dismutase(SOD) activity and glutathione(GSH) level effectively.The ROS scavenger N-acetyl-L-cysteine(NAC) had a similar inhibitory effect on melanogenesis.Conclusions:These results suggest that ginseng extract may inhibit melanogenesis by suppressing oxidative stress.
