卤代烷烃脱卤酶的芽胞表面展示技术研究

摘要
芥子气和α-卤代有机物有很强的细胞毒性,能对机体造成伤害,严重威胁到国家安全和公共安全。卤代烷烃脱卤酶(EC3.8.1.5)催化芥子气和α-卤代有机物水解具有作用快、条件温和、无腐蚀性等优点。但卤代烷烃脱卤酶在实际应用中仍面临一些技术难题,如对温度及pH变化敏感、贮存稳定性较差、酶的分离纯化造成生产成本增加等。芽胞表面展示技术能够保持展示蛋白在复杂条件下的生物活性,有效提升展示蛋白的贮存稳定性,而且构建的重组芽胞易收集,生产成本低廉,已在疫苗制备、生物催化等领域展现出良好的应用前景,目前尚未发现有关芽胞展示卤代烷烃脱卤酶的报道。本研究拟对枯草芽胞杆菌芽胞表面展示卤代烷烃脱卤酶的工作展开研究,以获得可在复杂环境下发挥催化作用、贮存稳定性高、生产成本低的功能芽胞。本论文的研究对于卤代烷烃脱卤酶的实际应用具有重要意义,对于防化领域和公共安全领域的发展也起到积极的推动作用。
Mustard gas and alpha halogenated harmed to human with a a strong cytotoxicity,then caused a serious threat to national security and public safety.Haloalkane dehalogenases(HLDs,EC3.8.1.5) could environmentally friendly and quickly decontaminate mustard gas and alpha halogenated with only nontoxic thiodiglycol produced.But HLDs is still facing some technical problems in the practical application,such as sensitive to temperature and pH,poorer storage stability,and high costs in separation and purification.Heterologous proteins displayed on Bacillus subtilis spore can keep its original structure and biological activity,then could be stored and used in different complex environments.Recombinant spores are safe and easy to collect with a low cost.In recent years,surface display of Bacillus subtilis spore has a great development and is applied to many fields,such as production of vaccines,drugs,enzymes and other functional protein.This study focus on surface display of Bacillus subtilis spore of HLDs,in order to obtain recombinant HLDs that had good catalytic activity in a complicated environment with high storage stability and low production cost.This study had great significance in practical application of HLDs,it also play a positive role in the development of chemical defense and public security.
