富锌滑子蘑菌丝体和子实体中半胱氨酸及其合成关键酶的研究

摘要
半胱氨酸在生物体内具有多种生理功能,也是合成谷胱甘肽的重要物质。本论文研究了不同富锌浓度对滑子蘑的菌丝体和子实体中半胱氨酸及半胱氨酸合成酶(CSase)的影响,培养浓度为:0、200、400、600、800、1000mg/kg。采用强酸性茚三酮检测酶促反应,通过亚硝酰铁氰化纳法测定半胱氨酸含量。实验结果:菌丝体和子实体中的CSase活性及半胱氨酸含量随着富锌浓度的增加整体呈上升趋势且均大于相对应的空白组;子实体中的CSase活性明显的高于菌丝体中的CSase活性,富锌浓度为1000 mg/kg时,菌丝体及子实体的CSase具有最大活性,分别是0.052U/mmol、0.986 U/mmol;子实体中的半耽氨酸含量在富锌浓度大于400mg/kg时高于菌丝体中的半胱氨酸含量,但富锌培养后滑子蘑的含量均大于相对应的空白组,在富锌浓度为1000 mg/kg时,菌丝体及子实体的半胱氨酸含量达到最大值,分别为250.0μg/mg、307.2μg/mg。进而得到结论,富锌对滑子蘑生长过程中丰胱氨酸的合成有明显的促进作用。
Cysteine in organism,has many physiological functions,but also an important material of synthesis of glutathione.This paper studied the effect of different concentration of Zn-enriched Pholita Namek mycelium and fruiting body of its cysteine and cysteine synthase(CSase),the training concentration as:0,200,400,600,800,1000 mg/kg.Using a strongly acidic ninhydrin method detected enzymatic reaction,the content of cysteine was measured by sodium nitroprusside method.The result:CSase activity and the content of cysteine in mycelium and fruiting body was in upward trend overall with the increasing concentration of zinc and each of them were greater than the corresponding blank group;CSase activity in fruiting body was significantly higher than that of mycelium,When the Zn-enriched concentration was 1000 mg/kg,the CSase activity in mycelium and fruiting body having maximum value,namely 0.052U/mmol,0.986 U/mmol;the content of cysteine in fruiting body was higher than its in mycelium at the Zn-enriched concentration greater than 400 mg/kg,but Zn-enriched Pholita Namek were greater than the corresponding blank group,in Zn-enriched concentration of 1000 mg/kg,the content of cysteine in mycelium and fruiting body reached a maximum,250.0μg/mg,307.2μg /mg.Then come to a conclusion,Zn-enriched on Pholita Namek growth process could have significant role in promoting about cysteine synthesis.
