碳纳米纤维/碳纤维固相微萃取联用LC-MS/MS检测植物激素

摘要
植物激素调控植物生长周期的每个生理过程,植物激素检测已成为近十几年来研究热点,尤其是活体原位分析领域。固相微萃取技术集采样萃取分离浓缩为一体,为实时检测植物激素,提供新思路。但该技术应用于植物样品分析中,有萃取纤维头易受污染,涂层容易脱落,价格也比较昂贵,富集率低下等缺点。本研究采用化学气相沉积法制备碳纳米纤维/碳纤维固相微萃取材料,建立一种浸提微富集实时检测植物激素的色谱分析方法。通过将碳纳米纤维修饰在碳纤维上,形成空间立体三维结构,有效的增加活性位点,进而提高富集率,解决浓度低难以实现实时检测的难题。该方法能够快速准确分析植物激素(JA,IAA,ABA),整个过程4 min即可完成,实现了植物激素的快速分析。3种目标物的回收率为72%~87%,RSD为3.30%~7.52%(n=6)。该方法对探究各类激素间的相互关系、认识激素调控植物适应环境的机制以及遗传育种等方面具有重要的指导意义。
Phytohormones play a vital role in plant growth and metabolic processes.It is usually rather difficult to determine phytohormones in plant tissues owing to their presence in trace amounts and to the coexistence of a wide variety of interferens.An effective new technique was developed to help carbon nanofibers(CNFs) grow on carbon fibers.The new solid phase microextraction(SPME) device manufactured with CF-CNFs has been tested with three different phytohormones(IAA,JA,ABA) in aqueous solutions,showing a great extraction efficiency with short adsorption time(3 min).The recoveries of 3 targets are 72%~87%,RSD are 3.30%~7.52%(n = 6).This new sorbent material can be then considered a suitable candidate for on-site,in-site and in-vivo extraction of analytes from biological samples.
