Isolation and Identification of Antagonistic Bacteria Against Soil-borne Pathogenic Fungus of Tobacco

This study is aimed to isolate and screen bacteria strains from tobacco rhizospheric soil,which have antagonistic activities against soil-borne pathogenic fungus of tobacco.Soil dilution plating technique and dual culture technique on agar plate were used for isolate and screen antagonistic bacteria.The antagonistic strains were identified by morphological features observation,physiological and biochemical experiments,and 16 S rDNA sequence analysis.30 rhizospheric soil were collected from tobacco filed in Luoning,Ruyang,Songxian and Yiyang countries in Luoyang,Henan province.180 bacterial isolates were obtained from 30 rhizosphere soil samples by separation and purification,and 49 strains have antagonistic function to the soil-born pathogenic fungi tested in this trial.Strains TS681、TS682、TS683were strongly antagonistic to three kinds of pathogenic fungi,and the inhibition zones to Pythium aphanidermatum,Phytophthora parasitica and Furasium oxysporum respectively more than 7.0 mm,10.0 mm and 9.0mm with the antagonistic rate respectively reaching above 51.4%,68.8%and 60.0%.All this three strains were identified as Paenibacillus polymyxa,and they may have the potential value of development and application for biological controlling of soil-borne fungi diseases of tobacco.
