Estrogen receptor gene susceptibility as possible risk factor in bone tissue abnormalities in adults exposed to fluoride in People's Republic of China

Objective:The aim of this study was to test the hypothesis that rs2941740 polymorphism and changes in DNA methylation of estrogen receptor alpha(ESRα) gene are involved in bone tissue abnormalities of adults living in fluorosis areas.Methods:A nested case control study was conducted among adults aged 18-55 years in Henan Province,China,based on the previous cross sectional study.ESRa gene rs2941740 polymorphism was identified with Taqman probe method.The methylation level in promoter region of ESRa gene was determined by Real-Time PCR.X-ray was used to detect changes of the bone tissue.Results:No significant association was found between the distribution of ESRa rs2941740 genotypes and the risk of bone tissue abnormalities.The methylation level in promoter region of ESRa gene in the case group was lower than that in the control group,but it was just observed in the male adults after stratified by gender.Furthermore,the methylation level was negatively associated with the urine fluoride concentration in male(r=-0.221,P=0.027).Unconditioned logistic regression analysis showed that after adjusted for age and gender,serum calcium and methylation status were protective factors of bone tissue,namely with the increase of serum calcium level and methylation level of ESRa gene,the risk of bone tissue abnormalities was decreased.It was not observed that the interaction between fluoride exposure and ESRa rs2941740 polymorphism on bone tissue.Conclusion:There was no significant association between rs2941740 polymorphism in ESRa gene and bone tissue abnormality in adults exposed to fluoride in drinking water.Bone metabolism may be related to the methylation level in promoter region of ESRa gene and affected by fluoride exposure.
