全细胞催化芦丁水解制备异槲皮素的研究

摘要
异槲皮素是广泛存在于药用植物、食品饮料、蔬菜水果中的天然黄酮类化合物,具有多种重要的生理活性及多方面优良的药理活性。目前其合成主要通过化学法和酶法进行。本研究探讨了基于全细胞生物催化的新型制备方法,通过催化芦丁水解制备获得了高纯度的异槲皮素。研究分析了全细胞催化剂培养制备过程中,不同的诱导条件对多种细菌和真菌细胞水解酶产酶活性的影响。研究结果表明,黑曲霉全细胞催化剂具有优异的催化活性和高区域选择性(99%);与纯酶法相比,全细胞催化法大大减少了繁琐的酶制备提纯工艺,降低了生产成本。本研究为异槲皮素及其相关结构化合物的高效制备提供了一条绿色、高效的新途径。
Isoquercetin is widely present in medicinal plants, food and beverage, natural flavonoids in fruits and vegetables, with pharmacological activity and physiological activity of a variety of important and excellent in many aspects. The new preparation method was discussed in this paper based on whole cell biocatalysis the catalytic hydrolysis of rutin, Isoquercitrin was obtained with high purity. Research and analysis of the whole cell catalyst to produce in the preparation process, different induction conditions of A variety of bacteria and fungi Cell effect of hydrolytic enzyme production activity. The results indicated that the Aspergillus niger whole cell catalyst has excellent catalytic activity and regional high selectivity(99%); compared with the method of pure enzyme, whole cell catalysis greatly reduces the complex of the enzyme, purification process, reduce the production cost. This study for Isoquercitrin and related structured complexes of high preparation provides a green and efficient new way.
