三倍体芍药品种花粉育性研究

摘要
为了充分了解芍药三倍体品种的育种潜力,促进我国芍药新品种培育工作的发展,本研究选取了3个芍药三倍体新品种‘Brightness',‘Roseletter'以及‘Coral Sunset',用I_2-KI染色法分别对其花粉活力进行了测定;并以国内二倍体品种‘粉玉奴'为母本,利用荧光显微技术,观察了三倍体品种在其柱头上的花粉萌发和花粉管生长情况。结果发现:①所观察的三倍体品种的花粉活力均非常低,其中品种‘Brightness'的花粉活力最高,为6.7%;品种‘Coral Sunset'的花粉活力最低,为2.2%。②所选三倍体品种的花粉在异种柱头上只有极少数的萌发,且花粉管极短,柱头乳突细胞及花粉管内出现大量胼胝质,未观察到花粉管伸入进花柱,同时发现大量花粉破裂及从柱头上脱落,组内远缘杂交亲和性非常差。
In order to fully understand the breeding potential of triploid herbaceous peony cultivars and promote the development of cultivating more new peony cultivars in China,3 triploid cultivars,' Brightness',' Roseletter' and ' Coral Sunset'were selected to investigate their pollen viability and fertility.Pollen viability was determined by I2-KI.Meanwhile,with the domestic diploid cultivar ' Fen Yu Nu' as female parent,the pollen germination and the pollen tube growth on its stigma were observed by fluorescence microscopy.The results showed:①The pollen viability of triploid cultivars were very low.Pollen viability of ' Brightness' was the best with 6.7%,while the ' Coral Sunset' s was the lowest with 2.2%.②The pollen grains of triploid cultivars had a very low germination rate on the maternal stigma.The pollen tubes were very short and couldn't enter the stigma,a lot of callose was found in papillose cells of stigma and pollen tubes.Meanwhile,a large number of pollen broke and detached from the stigma.The compatibility of distant hybridization was very low.
