团头鲂糖皮质激素受体基因的克隆及表达分析

为了了解糖皮质激素受体(GR)在团头鲂应激过程中的调控机制,本研究克隆分析了GR基因的序列特征,并运用实时荧光定量PCR技术检测了团头鲂GR的组织表达分布和在外源性皮质醇模拟应激下GR在组织中的表达变化。结果获得团头鲂GR1a、GR1b、GR2 mRNA序列分别包含了2199bp、2226bp、2241bp的开放阅读框,分别编码732、741、746个氨基酸。序列同源性分析及系统发育分析表明团头鲂GR均与鲤科鱼类GR聚为一支。组织分布的结果显示GR1在心脏、脾脏、卵巢以及皮肤中有较高的表达量;GR2在肝、下丘脑、垂体、肠道、精巢以及肌肉中表达丰度较高。应激状态下,下丘脑GR1在胁迫恢复的过程中存在表达先受到抑制再恢复正常的波动;而GR2在胁迫恢复早期均出现了上调表达,随后逐渐趋于稳定。垂体中,GR1的表达量出现了先升高后下降的趋势;而GR2也表现出先升高后降低的单峰型趋势。头肾中,受体的表达在胁迫恢复早期均表现出抑制现象。本研究为阐述GR的应激调控机理,深入了解团头鲂应激调控机制奠定了基础。
Glucocorticoid receptor(GR) plays important roles in stress. In the present study, we obtained the full length of M. amblycephala GR mRNA. The results showed that the full length of M. amblycephala GR1 a, GR1 b and GR2 mRNA containing a 2199, 2226, and 2241 bp open reading frame(ORF), and encoding 732, 741 and 746 amino acids, respectively. Sequence homology analysis and phylogenetic analysis showed that GRs were clustered with cyprinidae fish GRs. Tissue expression analysis showed that the higher expression of GR1 was observed in Heart,Spleen, Ovarian and Skin. While GR2 showed a higher expression in Liver, Hypothalamus,Pituitary, Gut, Testis and Muscle. The variation of GRs expression during the stress was detected after exogenous cortisol injection. In Hypothalamus, the changes of GR1 put up no clear variation tendency; while the changes of GR2 increasing slowly at the early stage and then gradually stabilized. In Pituitary, GR1 increasing at the early stage then decreasing at the later; the variation tendency of GR2 seems like a unimodal pattern. In Head kindey, all the receptors expression was decrease at the early stage.
