Human leukocyte antigen(HLA-G) is a crucial immunotolerant involved in malignant cells immune escape and disease progression.High frequencies of tumor cell membrane-bound HLA-G and increased peripheral soluble HLA-G(s HLAG) expression have been observed in various malignancies.In this study,the prognostic significance of plasma s HLA-G in 178 patients with colorectal cancer(CRC) was evaluated.Levels of s HLA-G were analyzed by specific enzyme-linked immunosorbent assay.Median of s HLA-G levels were significantly increased in patients compared with controls(36.8 U/ml vs 4.7 U/ml,p<0.001).Among CRC subgroups,Median of s HLA-G levels in died CRC patients were with significantly higher than that in the survived CRC patients(46.8 U/ml vs 27.44 U/ml,p=0.012).Moreover,patients with s HLAG above median levels(≥ 36.8 U/ml) had a significantly shorter survival time(p<0.001).Multivariate analysis revealed that,in addition to patient age and tumor status,s HLA-G could be an independent prognostic factor in CRC patients.However,no significant correlation was found between s HLA-G levels and clinical parameters of the CRC patients studied.Taken together,our study provided the findings that plasma s HLA-G level is a potent predictor for prognosis in colorectal cancer patients.
