鲍鱼生殖腺多糖构效关系的研究

本论文研究了鲍鱼性腺多糖(AGP)分别经酸水解及β-消去反应处理后,其结构以及其生物活性的变化。AGP经0.1mol/L HCl于100℃处理0~4h后,用NaOH调至中性,再用G-15凝胶柱脱盐后浓缩冻干。分别利用Spharose CL-6B和FT-IR分析AGP分子量与主要官能团的变化。结果表明,盐酸处理0.5h后,AGP发生明显降解,2h后水解趋于平稳。硫酸根含量明显降低,并有大量羟基生成。盐酸处理后,促淋巴细胞增殖、抗补体和促RAW264.7细胞增殖活性及抑制NO生成活性,但Hela细胞生长的抑制在1 h后增强,到4 h减弱。这四种活性均在酸处理0.5~1h呈现变化拐点,说明分子量的减少与含硫基团的缺失影响AGP的生物活性。采用β-消去反应结合Sevag法去除AGP中的蛋白,随着反应时间从0 h延长至72 h,共获得5个脱蛋白产物,其中蛋白含量逐渐减少,由原来6.48%降到1.68%,多糖相对含量则从55.62%升至65.9%,硫酸根含量变化较小,而糖醛酸含量则从21.25%升至27.03%。考察了β-消去产物对淋巴细胞增殖、抗补体活性、tBOOH致细胞氧化损伤以及Hela细胞增殖的影响。结果显示AGP脱蛋白产物的促淋巴细胞增殖活性及对tBOOH致细胞氧化应激损伤的保护活性均显著下降,蛋白含量越低,活性下降越明显。然而,抗补体活性和对Hela细胞增殖的抑制都有所提高。以上结果表明了分子量的减少、含硫基团的缺失和蛋白量都对AGP的生物活性有显著影响。本文结论有助于发展通过调整AGP蛋白量从而优化其生物活性,并且对AGP功能性食品的开发提供帮助。
Effects of dilute hydrochloric acid and β-elimination treatment on the structure and some bioactivities of abalone gonad polysaccharide(AGP) were evaluated.First,AGP was degraded by 0.1M HC1 at 100℃ for 0 ~4 h.After neutralization,the hydrolysate was desalted by sephadex G-15.Degradation degree of AGP was detected by HPLC equipped with TSKGEL G4000PW_(xl) and G2500PW_(xl) columns.Second,AGP was deproteinized by β-elimination reaction.Molecular size and main functional group were analyzed by Sepharose CL-6B chromatography and FT-IR,respectively.Significant degradation of AGP occurred after 0.5 h of acidic treatment,leveled off after 2 h.FTIR demonstrated obvious lost of sulfate groups and increased hydroxy groups.During acidic treatment,the lymphocyte proliferation,anti-complement,and NO-producing inhibition by RAW 264.7 cells bioactivities decreased significantly after 0.5 or 1 h,then rose again.But the inhibition effect on Hela cells growth increased after 1h,then decreased till 4h.All the above bioactivities demonstrated an inflection point after acidic treatment for 0.5- 1 h,suggesting molecular weight reduction and loss of sulfate groups influenced the bioactivities of AGP.After P-elimination reaction for 0 ~ 72 h,five fractions were obtained,with protein content reduced from the original level of 6.48%to 1.68%.Polysaccharide content of the fractions increased from 55.62%to 65.9%,21.25%to 27.03%for uronic acid content.Sulfated group content changed slightly.Deproteinization treatment showed negative effect on lymphocyte proliferation and protection function against tBOOH-induced oxidative damage,demonstrating a mode of lower protein content lower bioactivities.On the contrary,anti-complement activity and inhibition effect on Hela cells growth were enhanced.Above results suggested great influence of molecular weight reduction,sulfate groups loss and protein content on some bioactivities of AGP.It might be a useful strategy to control the protein content in AGP to satisfy the development of AGP function food with specific bioactivity.
