提取条件对麸皮阿拉伯木聚糖的得率及性质影响研究

阿拉伯木聚糖(AX)是小麦麸皮中一种重要的功能性非淀粉多糖,能够降低胆固醇、维持血糖水平和增强免疫力等,并且可在食品中作为稳定剂、增稠剂和乳化剂等,对人类健康具有积极的影响作用。目前,制备AX方法中主要以提取得率、纯度为指标确定提取工艺,而有关于AX相关性质随提取条件变化的影响研究未见报道。本研究采用碱酶法提取小麦麸皮中AX,研究提取温度、提取时间、NaOH浓度和料液比对AX得率的影响,并且利用离子色谱法、凝胶色谱法和分光光度计法分析产品单糖组分、相对分子量和阿魏酸含量在不同提取条件下的变化趋势,从而建立提取条件与相关性质之间的关系,提高小麦麸皮利用率。结果表明:随着提取时间的延长,提取温度、NaO)H浓度和料液比的增加,AX得率显著升高;当提取温度达到80℃,提取时间延长至3.5h,NaOH浓度为0.2mol/L,料液比为l:10时,得率达到4.92%,之后不再升高或升高缓慢。不同提取条件下所得的AX产品均主要含有阿拉伯糖(Ara)、木糖(Xly)、葡萄糖(Glu)和半乳糖(Gal)四种单糖;延长提取时间,提高反应温度、NaOH浓度和料液比,阿拉伯糖(Ara)和木糖(Xly)相对含量均增加,葡萄糖(Glu)相对含量下降,A/X比值相对增加,不同提取条件下A/X比值在0.68-1.05范围内;当提取温度为80℃,提取时间为3.5h,NaOH浓度为0.2mol/L,料液比为1:10时,阿拉伯糖(Ara)和木糖(Xly)相对含量均达到最大值。AX中阿魏酸含量随提取温度升高缓慢增加,当温度达到80℃时阿魏酸含量达到最高为3.88mg/g,随提取时间延长逐渐变小;NaOH浓度、料液比对阿魏酸含量影响较小。提取条件变化对AX相对分子量大小无明显影响,均集中于2200kDa和12-130kDa范围内;而对不同分子量区域的AX总糖浓度具有一定的影响:即高分子量总糖浓度随着提取时间,提取温度,NaOH浓度和料液比的增加而增加,直到提取温度80℃,提取时间3.5h,NaOH浓度0.2mol/L,料液比1:10时高分子量总糖浓度达到最大。综上,通过提取条件对小麦麸皮阿拉伯木聚糖的得率和性质影响研究表明:以AX得率为指标确定提取条件,既可保证AX得率又可获得富含阿魏酸和高相对分子量的AX产品。
Arabinoxylans are a type of important functional non-starch polysaccharides in wheat bran,which perform positive impacts on human health,such as reducing serum cholesterol,maintaining glycemic response and enhancing the immunity of humanbeings,as well as being used as stabilizer,thickener and emulsifier in food,etc..At present,the set up of AX preparation methods mainly accords to the extract yield and purity,howver,no researches about the effects of the changes of extraction conditions on the AX properties were reported.In order to estabalish the relationship between the extraction conditions and the AX properties as well as improve the utilization of wheat bran,AXs were extracted from wheat bran by using a alkali-enzyme method,and the effects of extraction temperature,extraction time,NaOH concentration and the ratio of material to liquid on the yield of AX were studied,and the monosaccharide components,the relative molecular weight and the content of ferulic acid colleted from different extraction conditions were analyzed by using ion chromatography,gel chromatography and spectrophotometer.,The results showed that with the increase of extraction time,extraction temperature,NaOH concentration and the ratio of material to liquid,the yield of AX increased significantly;when the extraction temperature reached 80℃,the extraction time was extended to 3.5h,the concentration of NaOH was 0.2mol/L,the ratio of material to liquid was 1:10 and the yield reached4.92%,after that,the yield of AX was not increased any more or just increased slowly.The mainly four types of monosaccharides of AX products collected from different extraction conditions were arabinose(Ara),xylose(Xly),glucose(Glu) and galactose(Gal).With the enhancing of the extraction time,reaction temperature,NaOH concentration and the ratio of material to liquid,the relative content of arabinose(Ara) and xylose(Xly) were increased,while glucose(Glu) was decreased.Under this condition,the A/X ratio was relative increase,and the A/X ratio under various extraction conditions were in the range of 0.68-1.05.When the extraction temperature was 80℃,the extraction time was 3.5h,NaOH concentration was0.2mol/L and the ratio of material to liquid was 1:10,the relative contents of arabinose(Ara) and xylose(Xly) reached the maximum value.The contents of ferulic acids in AX increased slowly with the enhancing of extraction temperature.When the extraction temperature reached 80℃,the content of ferulic acids reached a maximum value 3.88mg/g,and then became gradually smaller along with the extent of extraction time.The concentration of NaOH and the ratio of material to liquid had less influences on the content of ferulic acid.The changes of extraction conditions had no significant effects on the relative molecular weight of AX,which were centralized in the range of 2200 kDa and 12-130 kDa.However,The changes of extraction conditions showed certain effects on the total sugar concentration of AX in different molecular weight regions:the concentrations of total sugar in high molecular weight were increased along with the increasing of extraction time,extraction temperature,NaOH concentration and the ratio of material to liquid,and this concentration reached the maximum until the extraction temperature was80℃,extraction time was 3.5h,NaOH concentration was 0.2mol/L and the ratio of material to liquid was 1:10.In conclusion,the effects of the extraction conditions on the yiled and properties of wheat bran arabinoxylan illustrated that:AX yield could be used as an index to determine the AX extraction conditions,for the insurance both of the AX yield and AX products in high relative molecular weight riching in ferulic acids.
