Effects of recombinant human IL-10 on TNF-α、IL-1β、IL-17

Objective: To study the anti-inflammatory and immune mechanism of recombinant human interleukin-10( IL-10)in rats with adjuvant arthritis.Methods: Adjuvant arthritis model rats were divided into model group, IL-10 treatment group, methotrexate(Methotrexate, MTX) treatment group, IL-10 and MTX combination treatment group and normal control group, each group were given intraperitoneal injection. Immunohistochemical detection was conducted for synovial tissueexpression of tumor necrosis factor-α(TNF-α), IL-1β, IL-17; enzyme linked immuno sorbent detection was conducted for TNF-a, IL-1β, IL-17 levels in synovial fluids; RT-PCR was used for detection of relative expression levels of TNFα, IL-1β, IL-17 mRNAin synovial tissues.Results: The expression level of TNF-αin synovial tissues of joint in model group was 0.299±.031 ng/L,compared with 0.110±0.025 ng/L in normal control; IL-1βwas 0.233±0.015 ng/L in model group, compared with 0.021±0.005 ng/L in normal control; IL-17 was 0.358±0.014 ng/L in model group, it was 0.012±0.005 ng/L in normal control, The expression level of these three cytokines in model group were significantly higher than those in normal control group. Synovial fluid of TNF-a, IL-1β, IL-17 in IL-10 treatment group was 112.29±13.74, 55.90±9.69, 95.15±9.58ng/L respectively, and 150.73±18.96,85.76±11.36,155.65±12.51ng/Lrespectively in model group, there was statistically significat difference between the IL-10 treatment group and the model group(P < 0.05); Results for combination group were101.47±11.04,52.61±11.82,80.36±13.11ng/Lrespectively. Compared with the model group, the difference has statistical significance(P < 0.05). Relative expression ofsynovial tissue TNF-αmRNA, IL-1βmRNA and IL-17 mRNAin IL-10 treatment group was 2.53±0.1,2.35±0.1,1.98±0.1respectively, and in model group the results were5.23±0.1,3.06±0.1,3.05±0.1 respectively, the differences have statistical significance(P < 0.05); Results for combination group were 1.91±0.1,1.54±0.1,1.90±0.2respectively, compared with model group, the differences have statistical significance(P < 0.01).Conclusions: The anti-inflammatory and immune regulation effects of recombinant human IL-10 on adjuvant arthritis rats were induced by inhibiting the secretion and down-regulation of mRNA of inflammatory cytokines.
