聚维酮碘对不同黄鳝养殖水体的杀菌效果研究

聚维酮碘(PVPI)是水产养殖中的常用消毒剂。为确定黄鳝在室内外不同养殖方式下PVPI的合理用量,我们对不同养殖水体中PVPI的杀菌效果进行了研究。结果显示,25 ppm的PVPI对室外网箱养殖水(细菌浓度约10~3 CFU/ml)非常有效,12小时杀菌率达96.5%以上;而10 ppm的PVPI在相同条件下的杀菌率只有21.4%。在相同细菌浓度的室内养殖水体(过滤且高压灭菌的自来水)中,低至5 ppm的PVPI就足以杀灭所有嗜水气单胞菌。这一结果表明,水体中的有机质会明显影响PVPI的杀菌效果。为此我们利用富营养水(无菌的LB培养基)和非营养性水(无菌dd H2O)分别作为对照,结果显示,在10~3 CFU/ml细菌浓度下,要全部杀死水体中的细菌,PVPI的使用浓度分别为4000 ppm和1.25 ppm。同时我们测定了PVPI对黄鳝的半致死浓度,其24 h-LC_(50)为188ppm,且100 ppm作用24小时不会造成黄鳝的死亡。这些结果表明,在室外网箱养殖中,25-100 ppm的PVPI能够完全有效的对水体进行消毒而不造成黄鳝个体的损伤。
Povidone iodine(PVPI) is a common used disinfectants in aquaculture. In order to determine the appropriate dosage of PVPI to different rice field eel(Monopterus albus)aquaculture water,the bactericidal efficacy of PVPI on indoor laboratory aquaculture systems and outdoor net cage aquaculture systems were studied. Our results showed that 25 ppm of PVPI was very effective in outdoor aquaculture water(~10~3 CFU/ml) and more than 96.5 % bacteria were killed in 12 hours, however, it was only 21.4 % of bactericidal rate when PVPI was 10 ppm. In indoor aquaculture water with the same bacteria concentration, 5 ppm of PVPI was enough to kill all Aeromonas hydrophilia. 50 % lethal concentration of PVPI on rice field ell was investigated.The 24 h-LC_(50) is 188 ppm and 100 ppm of PVPI did not cause the death of the rice field eel in 24 hours. These results suggested that in outdoor aquaculture, 25-100 ppm of PVPI could disinfect water body effectively without any obvious harm to the individuals.
