贻贝蛋白的分离提取及性质研究

摘要
根据蛋白溶解性的差异,贻贝中的蛋白分为肌浆蛋白、肌原纤维蛋白和基质蛋白三种。三种蛋白在贻贝中的含量及性质报道较少,目前逐渐引起广大研究者的关注。本研究采用0.05mol/LpH=7的磷酸缓冲溶液浸提60min,提取制得肌浆蛋白;采用0.1moL/LpH=7含0.6mol/L氯化钠的磷酸缓冲溶液浸提30min,提取制得肌原纤维蛋白;采用3%的柠檬酸溶液70℃水浴浸提60min,提取制得基质蛋白;采用凯氏定氮法测定三种蛋白溶液的蛋白含量,十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)测定三种蛋白溶液的分子量分布。结果表明:肌浆蛋白占贻贝总蛋白的48.40%,肌原纤维蛋白占17.19%,基质蛋白占11.39%,三种蛋白总的提取率为76.98%。SDS-PAGE分析显示:肌浆蛋白条带较为广泛,其最主要组分的分子量约为45kDa;肌原纤维蛋白主要分子量在104kDa、45kDa;基质蛋白主要分子量在104kDa、89kDa、76kDa和14kDa。根据三种蛋白的特性可应用于不同的研究。
According to the different solubility of protein,Mytilus edulis contains sarcoplasmic protein,myofibrillar protein and myostromin.The report about content and property of three kinds of protein of Mytilus edulis is rare at present,but it draws researchers' attention gradually.The use of 0.05mol/L pH =7 phosphate butter solution to get sarcoplasmic protein after digestion of 60 min.And 0.lmol/ L pH =7 containing 0.6mol/L sodium chloride phosphate butter solution to gain myofibrillar protein after digestion of 30 min.Finally we got myostromin by using 3%citric acid solution and heating in70 t waterbath.We measured protein content of three kinds of protein by Kjeldahl method.And distinction of protein molecular weight was ensured by sodium dodecyl sulfate polyacrylamide gelelectrophoresis(SDS-PAGE).The results showed that three kinds of protein components were isolated from mussel representing 48.40%,17.19%and 11.39%of the total protein,respectively.The total protein extraction rate was 76.98%.At the same time,sarcoplasmic protein bands are wideand molecular weight of the most important fractionwas about 45kDa;Myofibrillar protein molecular weights were 104 kDa and 45kDa;myostromin molecular weights were 104 kDa,89kDa,76 kDa and 14 kDa.So according to the characteristics of the three kinds of protein,they can be used in different studies.
