LC-MS技术分析5种贝类中的含糖醛酸多糖

贝类中的含糖醛酸多糖具有多种生物学活性,是贝类的重要营养成分。为了解海洋可食用贝类中的含糖醛酸多糖种类,本文对强肋锥螺、纵带滩栖螺、纵肋织纹螺、单齿螺、海湾扇贝5种贝类的含糖醛酸多糖进行分析。采用木瓜蛋白酶和胰蛋白酶双酶法提取原料中的多糖,将所得多糖置于1.3M三氟乙酸(TFA)中在105℃下水解3h,然后采用1-苯基-3-甲基-5-吡唑啉酮柱前衍生化结合HPLC-MS/MS法分析水解产物中的二糖,通过与标准品保留时间对比及质谱数据解析发现,在纵肋织纹螺和单齿螺中主要含有硫酸软骨素(CS),纵带滩栖螺中含有→2)-α-Man(1→4)-β-GlcA(1→重复片段构成的非糖胺聚糖(AGSP),在强肋锥螺和海湾扇贝中存在CS和AGSP。此外,仅在强肋锥螺和海湾扇贝中发现少量肝素(HP),没有检测到透明质酸(HA)。为了获得更多的结构信息,将含有AGSP和CS的贝类多糖样品分别溶解于2M和0.2M TFA中在100℃下水解1h,然后采用PMP-HPLC-MS~n分析四糖与硫酸化/乙酰化的二糖,结果表明AGSP的结构为β-GlcA(1→2)-α-Man(1—4)-β-GlcA(1→2)-α-Man,强肋锥螺和海湾扇贝中的CS主要为C型。
Uronic acid-containing polysaccharides(UACPs) from shellfishes,an important nutrient,possess a variety of favorable biological activities.In order to understand the species of UACPs from marine edible shellfishes,the UACPs in five shellfishes,namely Turritella fortilirata Sowerby(TF),Batillaria zonalis(BZ),Nassarius variciferus(NV),Monodonta labio Linnaeus(ML),and Argopecten irradians Lamarck(AI) were characterized.The shellfishes were digested by trypsin and papain to obtain the polysaccharides.Then the polysaccharides were hydrolyzed by 1.3 M trifluoroacetic acid(TFA) at 105℃ for 3h,and HPLC-MS/MS upon l-phenyl-3-methyl-5-pyrazolone(PMP) derivatization was used to analyze the disaccharides produced from acid hydrolysis.The UACPs were identified by comparison of retention time(t_R) and MS data with references.Chondrotin sulfate(CS) was found as the major UACP in NV and ML,a non-glycosaminoglycan(AGSP) that was constructed by →2)-α-Man(1→4)-β-GlcA(1→ disaccharide repeats was the main UACP in BZ,and both of AGSP and CS were observed in AI and TF.In addition,heparin(HP) was only observed in trace amount in AI and TF,and no hyaluronic acid(HA) was found in these shellfishes.To obtain more structural information the polysaccharide samples which contain AGSP or CS from shellfishes were dissolved in 2 M TFA or 0.2 M TFA individually and heated at l00℃ for lh,then the tetrasaccharides and sulfated/acetylated disaccharides for analysis by PMP-HPLC-MS",and indicated that the structure of AGSP was β-GlcA(1→2)-α-Man(l→4)-β-GlcA(l→2)-α-Man,CS in AI and TF was Type C.
