超薄水滑石纳米片作为药物载体的研究

摘要
本文首先合成了单层结构的超薄水滑石纳米片(LDHs),并通过药物分子与LDHs层板的相互作用力,成功地将化疗药物DOX吸附于LDHs纳米片上,同时引入靶向分子FA,制备了具有可控释放能力的复合纳米粒子FA-DOX/LDH。计算得到DOX的负载量达到3.487 mg/mg(W/W)。采用KB细胞和L02细胞评价发现FA-DOX/LDH在具有优良肿瘤治疗性能,生物相容性以及细胞成像效果。因此,本工作合成的超薄水滑石纳米片具有高负载率,可明显降低化疗药物生物毒性,同时具备可控释放性能,在肿瘤治疗及成像领域具有广泛的应用前景。
Herein, we have reported a supramolecular FA-DOX/LDH fabricated by loading DOX to LDH single layer nanosheets through electrostatic interaction. Folic Acid(FA) was further introduced to achieve targeted cancer therapy. The loading amount of DOX in LDH single layer nanosheets is calculated to be 3.487 mg/mg(W/W). FA-DOX/LDH shows good targeted therapy performance and superior biocompatibility in vitro performed with KB and L02 cells. In addition, the excellent cellular imaging quality of FA-DOX/LDH is revealed by fluorescence microscopy images. Thus, the LDH single layer nanosheets exhibit high loading capacity as well as low cytotoxicity, which guarantees its practical applications as a controllable drug delivery vehicle in the fields of tumor therapy and cellular imaging.
