pH敏感的聚甲基丙烯酸空心纳米球用于药物释放和超声成像

摘要
本文利用硬模板法合成了pH敏感的聚甲基丙烯酸(HPMAA)空心纳米球,该纳米球具有均匀可控的空腔(约250nm)和壳层厚度(约20nm)。进一步用mPEG_(2000)-NH_2修饰增加其生物相容性,该HPMAA-mPEG空心纳球具有较小的细胞毒性、较高的药物负载率(DOX,52.4 wt%),DOX负载的空心纳米球可以被细胞内吞,并且在较高pH值条件下(7.4)有效释放药物。进一步将空心纳米球用于裸鼠活体内的超声成像表明该HPMAA-mPEG纳米球可被用作新型的超声成像造影剂。我们的实验结果表明,制备的HPMAA-mPEG空心纳球即可被用作pH敏感的药物释放运输也可以作为超声成像造影剂。
The pH-responsive hollow poly(methacrylic acid)(HPMAA) nanocapsules with uniform core size of ~ 250 nm and flexible shell thickness of-20 nm were synthesized by the hard-template protocol.After further surface modified the HPMAA with biocompatible mPEG_(2000)-NH_2,the HPMAA-mPEG nanocapsules displayed low cytotoxicity and high DOX-loading content(52.4 wt%).The DOX-loaded nanocapsules can be efficiently internalized by cells,and relase DOX under high pH(7.4).Then,the real-time in vivo ultrasound imaging of nude mice demonstrated that the HPMAA-mPEG nanocapsules can also be used as a novel ultrasound imaging contrast agent.Our restlts demonstrated the pH-responsive nanocapsules can be used as delivery vehicles of drugs and a novel ultrasound imaging contrast agent.
