Objective:d NK cells are generally considered to have a cytokine-secreting role rather than cytotoxic defence role of p NK cells.DSC,however,could recruit p NK cells to the decidua.We wondered whether DSC could change p NK cells phenotype towards d NK cells.Methods:We used FCM to test the expression of CD16 and ADAM-17 on p NK or d NK cells and DSC,respectively.ELISA kit tested the soluble CD16 in the coculture(p NK cells with DSC) supertanant or with ADAM-17 inhibitor,p NK conditioned medium or with ADAM-17 inhibitor.Results:(1)CD16+ d NK cells proportion was higher in abortional decidua than in normal.(2)ADAM-17+DSC proportion was lower in abortional decidua than in normal.(3)soluble CD16 concentration was higher in the coculture supertanant than in p NK and or with ADAM-17 inhibitor.(4)DSC regulated the shedding of CD16 on human p NK cells via ADAM-17.Conclusion:DSC may exert profound effect on the convention from pN K cells to dN K cells for a successfully pregnancy.
