基于7-羟基香豆素母体的过氧化氢荧光探针的合成研究

癌症,心血管疾病、神经退化性疾病等与人体衰退有关的疾病都和体内氧化应激过大有关,当活性氧簇(reactive oxygen species,ROS)等高活性分子在体内堆积时,就会引起氧化还原系统的失调,从而引发疾病。氧化应激过程主要副产物是过氧化氢(H2O2)。因此,能够适时准确地检测体内过氧化氢的含量具有十分重要的意义。本文主要介绍香豆素母环的6位为H(1a)、Cl(1b)、F(1c)的三种7-硼酸频哪醇酯香豆素的合成,并测定其与过氧化氢反应前后荧光性能的变化。研究结果表明1a、1b、1c合成产率依次降低,它们本身具有极微弱的荧光,加过氧化氢之后荧光性能大大增强。这三种探针表现出不同的荧光特征,可以作为细胞中过氧化氢浓度检测的有效荧光探针。
Cancer, cardiovascular disorders and neurodegenerative diseases have a close relation with large oxidative stress, which is the result of unregulated production of reactive oxygen species(ROS). The cellular mismanagement of oxidation-reduction chemistry will lead to illness. Hydrogen peroxide is a major ROS byproduct in living organisms and a common marker for oxidative stress. Thus it is very important to detect cellular hydrogen peroxide concentration timely and accurately. Three boronate esters of 7-hydroxycoumarin derivatives(1a, 1b, 1c) were synthesized and characterized, and the change in the fluorescence property upon reacting with hydrogen peroxide were measured. It showed that the synthesis of 1a has the largest overall yield while 1c has the least. The fluorescence was greatly enhanced after reacting with hydrogen peroxide for all three probes. The different fluorescence characterizations of these three 7-hydroxycoumarin derivatives make them effective fluorescent probes for the detection of hydrogen peroxide.
