RGD介导超顺磁性锌掺杂氧化铁纳米颗粒标记脑肿瘤癌细胞的磁共振成像研究

探讨RGD介导的荧光标记的超顺磁性锌掺杂氧化铁纳米颗粒(RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC)对于人脑肿瘤细胞U87的靶向性及磁共振成像(MRI)的可行性。制备Zn_(0.4)Fe_(2.6)O_4NPs-FITC探针及RGD靶向修饰的探针RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC,通过CCK-8试验验证其细胞毒性,并分别用Zn_(0.4)Fe_(2.6)O_4NPs-FITC(非靶向组)、RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC(靶向组)与人脑肿瘤细胞U87(1×106)共同培育0.5、1、2 h后做细胞悬液的MRI检查,测量横向弛豫时间(T2值)变化。采用激光共聚焦扫描显微镜观察纳米颗粒进入细胞内的情况。结果CCK-8比色试验培养液中不同浓度的RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC的细胞活性,各组间的差异无统计学意义。两种探针分别与U87细胞培育后0.5、1、2 h后MRI检查,两组细胞悬液的T2加权成像信号均下降,以2 h后更明显,靶向组在各时间段T2值下降的百分比均高于非靶向组。激光共聚焦扫描显示,两种标记的细胞内均可见纳米颗粒,靶向组中的颗粒数比非靶向组的更多。RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC对U87细胞有良好的靶向性,对人脑肿瘤细胞的早期诊断具有重大应用潜力。
To explore the feasibility of the human glioblastoma cell(U87) targeting and MRI abilities of the RGD mediated superparamagnetic iron oxide nanoparticles(RGD-Zn_(0.4)Fe_(2.6)O_4NPs-FITC).The cytotoxicity of RGD-Zn_(0.4)Fe_(2.6)O_4 NPs-FITC was verified through CCK-8 assay.Each group underwent MRI scan and the relative T2 values were recorded.Endocytosis of nanoparticles was observed through confocal laser scanning microscope.The cell viability of RGD-Zn_(0.4)Fe_(2.6)O_4 NPs-FITC in different concentrations showed no statistical difference according to the CCK-8 assay.The RGD targeting group showed a more significant decrease of relative T2 value at each time point.Cell imaging showed that the particles in RGD targeting group were more than that of the non-targeting group.RGD-Zn_(0.4)Fe_(2.6)O_4 NPs-FITC has favourable U87 cell targeting ability,and shows great potential in the early diagnosis of brain cancer.
