摘要
大防御素是一种具有广谱抗菌活性的抗菌肽,是参与生物先天性免疫的关键因子,具有杀灭病菌、保护机体的作用。本研究选取双壳类软体动物紫贻贝的大防御素成熟肽基因mMGBD作为研究对象,根据毕赤酵母的密码子偏爱性对该基因进行优化合成,将其与表达载体pPICZαA连接后电转至毕赤酵母(Pichia pastoris)X-33中,得到的阳性转化子在29℃、经0.75%甲醇诱导表达72小时后,通过SDS-PAGE分析证明重组大防御素rmMGBD得到成功表达。
Big defensin is a kind of antimicrobial peptide which possessesstrong activitiesagainstpathogenic bacteria. In the present study, we focused on the big defensinmature peptide from bivalve mollusk Mytilusgalloprovincialis. A codon-optimized m MGBD gene encoding big defensin mature peptide was synthesized and inserted into the pPICZαA expression vector,and recombinant expression vector pPICZαA-mMGBD was transformed into Pichia pastorisX-33 cells. The recombinant big defensin mature peptide(rm MGBD) was induced with1.0% methanol at 29℃ for 72 h. SDS-PAGE analysis demonstrated thatrm MGBD was expressed successfully.
