The swine streptococcosis was a great threat to China pig industry,Streptococcus equi.ssp.zooepidemicus(SEZ) was one of the major pathogens.SEZ ATCC35246 was a classical virulence strain,while SEZ ST171 was an artificial attenuated vaccine strain in China.In this study,we employed the SILAC and LC/MS technology to find out the differentiation in macrophage between these two bacterial infection.87 upregulatedproteins and 135 downregulated proteins were identified.We chose 10 genes for the Real-time PCR and 3 proteins for the Western blot to verify the proteomic results.AU different expression genes were analyzed with R package "clusterProfiler" to get their GO and KEGG informations.Results showed that some of the down regulated genes were associated with immunity functions and up regulated gene were related to cytomembrane and cytoskeleton regulation.Raw264.7 cells phagocytosis rate and cytokine genes transcription to SEZ ATCC35246 and ST171 infection were detected in order to prove bioinformatics conclusion.These results illustrated the different influence to macrophage phagocytosis and cytokine expression might be important reasons of different infection phenotype of SEZ ATCC35246 and ST171.This research willexplainthe mechanism of host immunity responses to SEZ ST171 and how SEZ ATCC35246 invading phagocytosis,provide information on potential therapy and vaccine development targets.
