八钼氧酸盐复合材料去除人血浆中高丰度蛋白

通过静电作用将多金属氧簇八钼氧酸盐[(C_(16)H_(36)BrN)_4Mo_8O_(26),(Mo_8O_(26))]与多孔金属有机骨架MIL-101(Cr)相结合,制备了新型的复合材料Mo_8O_(26)@MIL-101。采用XRD、TG、FT-IR、Raman、SEM、EDS和BET等技术手段对该材料进行了表征,所得材料性质稳定,比表面积高达1499m~2/g。基于Mo_8O_(26)~(4-)与组氨酸之间的共价配位作用,该复合材料对高丰度组氨酸蛋白HSA和IgG表现出良好的吸附选择性。在pH=5.0的条件下,对HSA和IgG的吸附容量分别为785.6 mg/g和420.1 mg/g。据此建立了固相吸附过程用于蛋白质的吸附与去除。SDS-PAGE(Fig.1)和2D-PAGE(Fig.2)电泳结果表明,人血浆样品经Mo_8O_(26)@MIL-101吸附处理后,高丰度HSA和IgG得到有效去除;去除后的血浆样品经LC-MS/MS可鉴定出334种血浆蛋白,其中中低丰度蛋白占蛋白总数的53.66%。
A novel composite,namely Mo_8O_(26)@MIL-101,is prepared by the electrostatic binding of octamolybdate(Mo_8O_(26)) with the porous Metal organic framework MIL-101(Cr).The obtained composite owns a surface area of 1499m~2/g.Due to the coordination between the Mo_8O_(26)~(4-) and histidine,the composite exhibits excellent adsorption towards HSA and IgG.At pH=5.0,the adsorption capacities for HSA and IgG is 785.6 mg g~(-1)and 420.1 mg g~(-1).SDS-PAGE and 2D-PAGE results indicate that efficient removal of abundant HSA and IgG from plasma is achieved.A total of 334 plasma proteins could be identified from the treated plasma by LC-MS/MS and the percentage of proteins of middle and low abundance is 53.66%.
