A comparative study of the pathogenicity of two different Eimeria tenella isolates in China

Introduction Avian Coccidiosis caused by Eimeria species is recognized as the parasitic disease with the greatest economic impact on poultry industries worldwide.Eimeria tenella,one of the seven species of protozoan parasites,is the most pathogenic species that cause cecal haemorrhage.To investigate the virulence between different E.tenella isolates(or strains) and screen vaccine candidate strain,a comparison of the pathogenicity of two E.tenella isolates from the Hebei and Xinjiang province of China was evaluated in specific pathogen free(SPF) chicken.Material & Methods Seventy 25-day-old SPF chickens were randomly assigned to two isolates.Each isolates with 6 groups of 5 chickens were inoculated with 5×10~2,1×10~3,5×10~3,1×10~4,2.5×10~4and 5×10~4 sporulated oocysts,respectively.Another ten were used as a non-infected control.All surviving birds were weighed on the 5.5th day postinoculation and were necropsied.In order to further research the pathogenicity of Zhangjiakou isolate,forty eight 25-day-old SPF chickens were infected with 5×10~3,2.5×10~4,5×10~4,1×10~5,2.5×10~5,5×10~5,1×10~6,and 0 sporulated oocysts,respectively.An assessment of the severity of the infection was based on mortality,cecal lesion scores,and survivor weight gain.Results 60%(3/5) chickens inoculated with 5×10~4 oocysts of Xinjiang isolate died 5 days after inoculation,20%(1/5) in the group of 1×10~4 on 6 days.However,all chickens infected with Zhangjiakou isolate were alive even the inoculation was 1×10~6 sporulated oocysts.Compared with the non-infected control,no significant difference in mean body weight was observed in all Zhangjiakou isolate inoculated groups,but significant difference appeared in the groups of 1×10~4,2.5×10~4 and 5×10~4 of Xinjiang isolate.Only a few scattered petechiae were seen on the cecal mucosa even in the seriously infected groups.However,in the groups of Xinjiang isolate,the mean body weight was decreased gradually with the increasing inoculation dosage.Cecum bleeding is severe,with clotting appearing in the distal end of the pouch with only 5×10~2 oocysts infection of Xinjiang isolate.Conclusion The results implied that different isolates(or strains) of E.tenella displayed different pathogenicity.Although further researches should be carried out in other biological characteristics,Zhangjiakou isolate with such low virulence has potential use in live vaccine development.
