Precocious line selection of Eimeria tenella isolated from Hebei, China

Introduction Avian coccidiosis, a protozoan disease, which caused by Eimeria species is considered of worldwide economically important disease for poultry farm. Given the drug resistance, vaccination with attenuated lines of the parasites is rapidly gaining great prominence in commercial poultry production. Attenuated vaccine lines of Eimeria have been developed widely in Britain, USA and Spain. Until recently, however, there have been no attenuated coccidiosis vaccines available in China. E. tenella capable of causing haemorrhage in the caecum of the fowl is now recognized as a significant pathogen. All the facts have stimulated the selection of precocious line of E. tenella, which will be more efficient for preventing coccidiosis in China. Material & methods All equipment and facilities including isolators and cages were cleaned and sterilised using ammonia for at least one night before use. All feed was formulated with no coccidiostat. Feed and water were stored at 80°C for 2 hours at least to avoid contamination. The original parent line of E. tenella, came from Hebei province of China, was cultivated from a single oocyst and the pathogenicity assay was conducted. The precocious line was obtained by repeatedly selecting early development of oocysts. Each passage was propagated by orally administering oocysts to coccidia-free chickens, and the prepatent period was detected. Oocysts were harvested from faeces a few hours later. Whenever possible, the selection pressure was applied during selection for precociousness. The mean dimensions of sporulated oocysts of the 10 th generation and parent strains were measured to determine if selection for precociousness resulted in a change in oocyst size. 100 randomly encountered oocysts were measured on the photographs pictured by microscope. Results The parent line was identified as a low virulence strain because no bird died even the inoculation dose was 1×106 sporulated oocysts. Selection for precociousness was effective in reducing the prepatent period by 11 hours through 14 generations. What's more, a significant difference in the oocysts dimensions between parent line and 10 generations of selection appeared. The mean size of sporulated oocysts of the parent line and 10 th generations of the precocious line are 22.42×17.79 and 20.14×16.76, respectively. Conclusion The results show that after pressure selection the line in the present study has possessed precocious characters. However, the selection of precocious line should be continued to obtain genetically stable strain.
