分层级三维介孔硅基材料用于生物标志物检测

摘要
临床样本的生物标志物的提取效率高度依赖材料的比表面积和孔道的通量体积~[1]。介孔纳米粒子和二维介孔硅基材料在提取生物标志物的过程中,由于它们的比表面积、通量体积比较小,很容易被血清/血浆中丰富的蛋白质和脂质堵住孔道,这样会大大抑制吸附效率。当前常规三维介孔硅基材料制备工艺相对复杂,在临床检测应用过程中,实际操作的灵活性稍微偏低。因此运用新的方法,新的工艺来构筑功能性分层级三维介孔硅基材料已经是十分迫切的,而且这种功能性材料用于生物标志物的精准诊断富有创新性和挑战性~[2]。这种功能性材料由于拥有更大的比表面积和更高的通量体积,能够避免蛋白质和脂质的堵塞,会很大程度上提高生物标志物的吸附效率,这为临床样本检测可能提供了一种新的策略。本研究旨在通过调控软模板的种类和硬模板的尺寸来搭建不同的孔道结构的硅基材料,以进一步深入应用到临床样品的检测中。该研究可以为临床样品检测提供新的策略,具有巨大的生物应用价值。
The extraction efficiency of biomarker from clinical samples is highly dependent on the surface area and throughput volume of channel.When mesoporous nanoparticles and two-dimensional mesoporous silica materials apply in the process of extraction of biomarkers,due to their specific surface area and throughput volume are small,their channel is easy to be blocked by the protein and lipid of serum/plasma,it will greatly inhibit the adsorption efficiency.Currently,It is relatively complicated for the preparation process of conventional three-dimensional mesoporous silica materials,and is slightly lower in the flexibility of actual operation.Therefore functional material with larger specific surface area and higher throughput volume,which can be avoided the blockage of protein and lipid,will largely improve the adsorption efficiency of biological markers,and may provide a new strategy for the clinical samples.This study aims to build functional silica materials which apply in the detection of clinical samples.
