Microarray Analysis on Differentially Expressed Genes Associated with Wheat Lr39/41 Resistance to Puccinia Triticina

Wheat leaf rust,caused by Puccinia triticina(Pt),is one of the most severe fungal diseases threatening global wheat production.Utilization of leaf rust resistance(Lr) genes is the major solution for this issue.Wheat isogenic lines carrying Lr39/41 gene still showed a moderate or higher resistance to most of the leaf rust races detected in China.In our study,a typical hypersensitive response(HR) has been observed by microcopy in Lr39/41 isogenic lines inoculated with avirulent Pt race from24 hours post inoculation,indicating a possible NBS-LRR and program cell death-related pathway for the mechanism of Lr39/41 gene.Two Lr39/41 resistance-associated suppression subtractive hybridization(SSH) libraries have been established.One with 3 456 clones constructed between Pt-inoculated and non-inoculated Lr39/41 isogenic lines.Another with 2 544 clones constructed between Pi-inoculated Lr39/41 isogenic lines and Thatcher susceptible lines.Microarray hybridization has been carried out on the established SSH libraries with RNAs extracted from Pi-inoculated and non-inoculated Lr39/41 isogenic lines and Thatcher susceptible lines,respectively.Differentially expressed genes(DEGs) were analyzed by significance analysis of microarrays(SAM) and a total of 386 clones with significant induction or suppression were further sequenced.Sequences were then annotated with GenBanknr database,URGI wheat genome database and gene ontology(GO) database.In total of 36 Lr39/41 resistance-related DEGs were found,many of which were previously reported in plant defense response.Four Lr39/41 resistance-related DEGs were also detected as Thatcher susceptible-related DEGs,indicating a potential involvement of basal resistance for these genes.Relative expression levels of 12 selected Lr39/41 resistanceassociated DEGs during Pt infection were further validated by qRT-PCR assay.A brief profile of DEGs associated with Lr39/41 resistance had been drafted.
