A review of the pharmacological mechanism of Pseudolaric acid B and its derivatives

Considering that extensive immunosuppressive agents have adverse reaction on normal cells to restrain their clinical application, researchers are devoting to find some novel agents with less side-effects and more security from Traditional Chinese Medicine(TCM). Cortex pseudolaricis, a kind of traditional Chinese medicine, has been used to treat tinea or other skin diseases since long time ago. Pseudolaric acid B(PAB) is the main effective diterpenoid of Cortex pseudolaricis. According to previous studies, PAB has been proved to markedly inhibit the growth and proliferation of many tumor cell lines by inducing mitotic catastrophe and apoptosis, and affect cell cycle, which makes it as a novel microtubule inhibitor. Moreover, it has been reported to exert favorable anti-inflammatory and immunosuppressive activities with less cytotoxicity.Through analyzing the correlational studies in recent decades, we have made a review on PAB and its derivatives, espe-cially the research development of its pharmacological activities.Even though the experimental utility of PAB is prevailing accepted, the mechanism is not completely clear so that we have a long way to explore the potential mechanisms of PAB against tumor and immune-related diseases.
