液相色谱质谱法测定洗发水中去屑因子吡硫翁锌和甘宝素在头皮上的沉积量

摘要
使用去屑洗发水后,检测头皮上沉积的去屑因子的量,可以帮助判断去屑效果。本文建立了测定洗发水中去屑成分吡硫翁锌(ZnPT)和甘宝素(CBZ)在头皮上的沉积量的液相色谱质谱方法。缓冲液清洗(buffer scrub)作为临床取样方法,在受试者使用测试用去屑洗发水后,采集头皮样品。ZnPT经过衍生化和CBZ同时被液相色谱质谱法定量。该方法可以测定头皮上纳克级的ZnPT和CBZ。采用这个方法,成功测定了在使用单去屑因子(ZnPT)和双去屑因子(ZnPT和CBZ)洗发水后不同时间点,头皮上ZnPT和CBZ的沉积量。
The measurement of anti-dandruff(AD) actives deposited onto the scalp from AD shampoos is helpful to evaluate the AD efficacy.A sensitive ultrahigh performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS) method has been developed and validated for simultaneous quantification of zinc pyrithione(ZnPT) and climbazole(CBZ) deposited onto human scalp from AD shampoos.Scrubbing with a buffer solution was used as the sampling method for the extraction of ZnPT and CBZ from scalp.Derivatization of ZnPT was carried out prior to UHPLC-MS/MS analysis.The limit of detection for ZnPT and CBZ was established to be 1 and 2 ng/mL,respectively.This sensitivity enables the quantification of ZnPT and CBZ at deposition levels in the low ng/cm2 range.The method was successfully applied for the analysis of scalp buffer scrub samples from an in vivo study.The levels of ZnPT and CBZ deposited on the scalp at different time points after application of the AD shampoo were measured.
