含硫香料降低金黄色葡萄球菌毒力基因nuc的表达

含硫香料对致病菌毒力基因表达的影响还没有被研究,本文选取了异硫氰酸苄酯(benzyl isothiocyanate,BNITC)和二烯丙基二硫醚(diallyl disulfide,DADS)两种含硫香料,研究BNITC和DADS对金黄色葡萄球菌耐热核酸酶基因(nuc)表达的影响。通过微量肉汤稀释法测定BNITC和DADS对金黄色葡萄球菌的最低抑菌浓度,并根据OD_(600nm)值绘制生长曲线,通过实时定量RT-PCR方法考察亚抑菌浓度下的BNITC和DADS对金黄色葡萄球菌nuc基因表达的影响。结果显示,BNITC和DADS对金黄色葡萄球菌ATCC6538的亚抑菌浓度分别是0.61 mmol/L和310.25 mmol/L。BNITC的浓度由1/4MIC、1/8MIC、1/16MIC、1/32MIC变化至1/64MIC、1/128MIC,与对照组相比,nuc基因的相对表达量分别下降至12.41%、34.72%、38.39%、47.99%、59.63%和89.76%。同样DADS在其亚抑菌浓度的条件下,也明显抑制了nuc基因的表达。以上数据表明,亚抑菌浓度下的BNITC和DADS会减少金黄色葡萄球菌毒力基因nuc的表达,且这种抑制作用呈现一定的浓度依赖性。因此,以BNITC和DADS为代表的含硫香料可能对金黄色葡萄球菌耐热核酸酶基因所引起的感染起到一定的削弱作用,并防止葡萄球菌的食物中毒。
The influence of sulfur-containing flavors on the virulence factor expression of pathogens has not been elucidated,This article selected two kinds of sulfur-containing flavors,that is benzyl isothiocyanate(BNITC) and di-allyl disulfide(DADS),The aim of this study was to explore the effect of BNITC and DADS on the thermostable nuclease(nuc) production of Staphylococcus aureus.The minimum inhibitory concentration(MIC) of BNITC and DADS against S.aureus was determined using a broth microdilution method,and growth curve based on OD_(600nm) value,real-time quantitative reverse transcriptase-polymerase chain reaction were performed to evaluate the effect ofsubinhibitory concentrations of BNITC and DADS on the virulence factor nuc of S.aureus.The result showed th-at MIC of BNITC and DADS against S.aureus ATCC6538 was found to be 0.61 mmol/L and 310.25 mmol/L re-spectively.Treatment of BNITC at subinhibitory concentrations ranging from1/4MIC、1/8MIC、1/16MIC、1/32 MICto 1/64MIC、1/128 MIC,by comparing control group,relative expression of the nuc were decreased to 12.41%、34.72%、38.39%、47.99%、59.63%and 89.76%respectively.Subinhibitory concentrations of DADS also inhibited the expression of thenuc obviously.These results suggest that subinhibitory concentrations of BNITC and DADS redu-ces the expression of virulence factor nuc by S.aureus.And a dose-dependent attenuation of themostable nuclease(nuc) production was observed in S.aureus.Therefore,benzyl isothiocyanate(BNITC) and diallyl disulfide(DADS) may be useful in the attenuation of nuc producing S.aureus infections,and to prevent staphylococcal food poison-ing.
