干酪乳杆菌调节Th17和Treg分化减轻鼠肠道炎症

为探讨干酪乳杆菌对动物细菌性肠炎的作用机制,18只Balb/c小鼠被随机分成3组,肠炎组:用2.0×10~8 cfu/ml的ETEC K88 0.2 ml连续灌胃3 d;干预组:用2.0×108cfu/mL的L.casei给鼠连续灌胃7 d,然后用2.0×10~8 cfu/ml的ETEC K88 0.2 ml连续灌胃3 d;对照组,用生理盐水灌胃3 d。利用ELISA方法检测不同处理组小鼠外周血和十二指肠IL-6、TGF-β的含量,流式细胞术检测肠系膜淋巴结和脾脏中CD4~+CD25~+Foxp3+Tregs和CD4~+IL-17A~+Th17细胞数量,荧光定量PCR及Western blot方法检测肠系膜淋巴结中Foxp3和RORγtmRNA及蛋白含量,并观察十二指肠的病理组织学变化。结果表明,干预组小鼠外周血和十二指肠的IL-6含量明显低于肠炎组,且差异显著(p<0.05);而TGF-β含量明显提高,且差异显著(p<0.05)。与肠炎组相比,干预组小鼠脾和肠系膜淋巴结中CD4~+CD25~+Foxp3+Tregs的百分含量被明显提高(p<0.05),CD4~+IL-17A~+Th17的百分含量明显降低(p<0.05)。干预组Foxp3mRNA及蛋白的表达量高于肠炎组,而RORγt的mRNA及蛋白的表达量低于肠炎组,且差异显著(P<0.05)。经L.casei干预肠炎鼠后,十二指肠炎症明显减轻。证明干酪乳杆菌有可能通过调节Th17/Treg的平衡而减轻鼠肠道炎症。
In order to study the ability of Lactobacillus casei to ameliorate murine enteritis,Mice numbering 18 individuals were randomly divided into the enteritis group(administered 0.2 mL of 2.0×10~8 cfu/ml ETEC K88 daily for 3d);the intervention group(administered 2.0×10~8 cfu/ml L.casei for 7 d,then 2.0×10~8 cfu/ml ETEC K88 for 3 d) and the control group(with physiological saline).IL-6 and TGF-β content in mouse peripheral blood and duodenum was detected by ELISA;the numbers of CD4~+CD25~+Foxp3~+ T-regulatory cells(Tregs) and CD4~+IL-17A~+ Th17 cells in the mesenteric lymph nodes(MLN) and spleen were detected by flow cytometry,and quantitative reverse transcription PCR and western blot analysis were used to measure Foxp3 and RORyt(retinoid-related orphan receptor-y) mRNA and protein expression in the MLN.Furthermore,histological changes in the duodenum were observed.The results indicated that for the intervention group,IL-6 content in mouse peripheral blood and duodenum was significantly lower than in the enteritis group(p<0.05),while TGF-β content was significantly increased compared to the enteritis group(p<0.05).For the intervention group,the percentages of CD4~+CD25~+Foxp3~+ Tregs in spleen and MLN were increased(P<0.05),while the percentages of CD4~+IL-17A~+ Th17 cells were decreased compared to the enteritis group(P<0.05).The expression of Foxp3 mRNA and protein in the intervention group was higher than in the enteritis group,while RORyt mRNA and protein were significantly lower(p<0.05).After mice in the enteritis group were treated with L.casei,duodenal inflammation was relieved.This study demonstrated that L.casei could have possible implications for the ETEC induced intestinal inflammation by regulating the ratio imbalance of Th17/Treg cells.
