低温胁迫对尼罗罗非鱼生物学变化的影响

为了探究8℃低温胁迫下尼罗罗非鱼身体失去平衡的原因,实验选取了尼罗罗非鱼和草鱼作为研究对象。设定28℃为对照组和8℃低温处理为实验组,每组设定6条鱼。测定了血清指标天冬氨酸氨基转移酶AST、谷丙转氨酶ALT、尿素UREA、尿酸UA、葡萄糖GLU、乳酸脱氢酶LDH、甘油三酯TG,及TUNEL法检测鳃的凋亡情况。结果表明:罗非鱼与对照组相比,实验组AST,UA,TG水平在低温胁迫6h显著性升高(p<0.05),ALT水平也显著性升高(p<0.01),GLU,LDH水平也有升高,但是没有显著性差异。草鱼的AST,ALT,GLU水平实验组与对照组比虽然均出现升高,但是差异不明显,UREA,UA,LDH,TG水平变化不明显。TUNEL法检测到罗非鱼鳃细胞在低温处理6h已结出现了凋亡信号,而草鱼未出现凋亡信号。因此,低温处理下罗非鱼身体失去平衡,可能与肾脏受损和鳃出现凋亡相关。
In order to the reason of the Tilapia lose body equilibrium when water temperature declined to 8 ℃,tilapia(Oreochromisniloticus),andgrass carp(Ctenopharyngodon- idellus) were selected as research object. We set up two groups: a control group(28 ℃) and a treatment group(8 ℃),six fish in every group. The lever of aspartate transaminase(AST),alanine aminotransferase(ALT),UREA,UA,GLU,LDH and TG were measured.The apoptosis of gill were detected by TUNEL. Results showed that compared with the control group of Tilapia,group,the treatment group had the following serum parameters significantly increasedat 6h:AST,UA and TG(p<0.05),ALT(P<0.01),the lever of GLU and LDH on significant differences.The treatment group of lever of AST,ALT and GLU of grass carp were higher than the control group. The lever of UREA,UA,LDH and TG were not obvious. Apoptotic cells were detected in gills of Tilapia exposed cold temperature at 6h by TUNEL, but the grass carp were not detected. Therefore, the Tilapia lose body equilibrium when water temperature declined to 8 ℃, was related with kidney damage and apoptosis of gill.
