目的探讨表面解析常压化学电离质谱(SDAPCI-MS)检测大肠埃希菌(E.coli)的挥发性有机代谢产物(VOCs)对快速鉴定E.coli的临床价值。方法 BACT/ALERT 3D全自动血培养仪进行增菌培养。VITEK2-compact全自动微生物分析仪进行菌种鉴定。SDAPCI-MS检测菌落上层气体和血培养瓶上层气体VOCs。结果 m/z 116特异地仅存于E.coli中,且其信号强度与培养时间和细菌初始浓度成正比。M/z 116经二级质谱确定为吲哚。结论 VOCs的快速SDAPCI-MS检测,操作简便、重复性高、鉴定结果可靠,生物标志物吲哚尤为适用于临床E.coli的鉴定。
Objective The investigation on volatile organic compounds(VOCs) of common pathogens by Surface analytical atmospheric pressure chemical ionization mass spectrometry(SDAPCI-MS) analysis for the clinical value of E.coli identification. Methods BACT/ALERT 3D automated blood cultures instrument enriched the bacteria. Identified by VITEK2-compact automated microbial analyze. VOCs of solid cultures and blood culture were detected by SDAPCI-MS. Results m/z 116 specifically only found in E.coli, and the signal strength was proportional to the incubation time and the initial concentration of bacteria. M/z 116 was determined by tandem mass spectrometry as indole. Conclusion Fast SDAPCI-MS detection of VOCs, is an easy operation, with high repeatability, reliable identification results, biomarkers indole is particularly suitable for the identification of clinical E.coli.
