The Abortion Mechanism and Fertility Restoration of Microspores during the Anther Development in Cotton Genic Male Sterile Line Yu 98-8A

Although cotton genic male sterility(GMS) plays an important role in the utilization of hybrid vigor,its precise molecular mechanism remains unclear.To characterize the molecular events of microspore abortion and fertility restoration,transcriptome,microRNA expression analysis combined with histological observations and hormones treatments were conducted in the cotton GMS line Yu 98-8A.Histological analysis showed the shriveled tetrad was the first significant microscopic phenotype that developed,indicating that the pollen mother cell formation phase and the following meiotic cycle are vital periods of microspores abortion in MS of Yu 98-8A.Biochemical transcriptome bioinformatic analysis indicated that the differentially expressed genes(DEGs) mainly involved in sugar and starch metabolism,oxidative phosphorylation,and phytohormones might play a critical and complicated role in pollen abortion.At the same time,microRNA expression analysis also showed indole-3-acetic acid(IAA) and gibberellic acid(GA) signalling tranduction pathways may play pivotal regulatory function in male sterility.Further studies have shown that regulating endogenous GA3 content in young buds restore the fertility of Yu 98-8A effectively.These results provide an important strategy of overcoming the bottleneck in cotton heterosis utilization for us to avoid the disadvantages(removal of about 50% fertile plants) of cotton genic male sterile lines effectively.
