颤藻对对虾抗氧化酶浓度及免疫相关基因表达的影响

颤藻是对虾养殖池塘常见的有害微藻。为了研究颤藻对对虾抗氧化系统及免疫系统的影响,本研究分析了凡纳滨对虾在注射半致死浓度的绿色颤藻细胞粗提液后,肝胰腺组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽(GSH)等抗氧化酶浓度的变化。同时通过实时定量PCR(Real-time PCR)技术检测了LvToll1、LvToll2、hsp90和Lzm等免疫相关基因的表达情况。研究发现,对虾注射粗提液后,体内SOD、CAT、GSH浓度均显著升高(P<0.05),其最高浓度分别是实验初始水平的2.3倍、11.5倍和1.7倍。受粗提液刺激后,对虾LvToll1、LvToll2和hsp90等基因的表达量有不同程度的上调,其中LvToll1、LvToll2和hsp90最高表达量分别是初始水平的40倍、1.5倍和4.6倍,Lzm基因的表达量在整个试验期间都低于初始水平,最低值只有初始水平的0.09倍。研究表明,颤藻细胞粗提液可能刺激对虾产生大量自由基,从而增加抗氧化酶蛋白表达,同时刺激对虾免疫相关基因作出不同的响应。
In order to explore the effect of Oscillatoria on antioxidant system and immune system of shrimp, the study collected hepatopancreas tissue at experimental time point after Litopenaeus Vannamei injected with Oscillatoria chlorine cell crude extract of half lethal concentration, and traced the concentration change of the antioxidant enzymes, SOD, CAT, and GSH, and the expression of immune genes, LvToll1, LvToll2, hsp90 and Lzm by Real-time PCR. The results showed that the concentration of SOD, CAT, and GSH obviously increased, the top level of which was 2.3 times, 11.5 times and 1.7 times higher than the initial level, respectively. And the expression level of LvToll1, LvToll2 and hsp90 was also up-regulated, which increased 40 times,1.5 times and 4.6 times in most of the time, but the expression level of Lzm was lower than initial level during the whole experiment. The floor level was only nearly nine percent of initial expression level. The results demonstrated that the crude extract may induce shrimp to produce massive free radical, which increase the protein expression of antioxidant enzyme.
