锦鲤精原干细胞的种间移植

摘要
生殖细胞移植技术是指将供体动物的生殖干细胞通过细胞移植技术移植到受体动物的性原基,移植细胞随后在受体动物性腺内增殖、发育分化,最终生成供体来源的成熟配子并繁育后代。这项技术的发展与应用促进了鱼类的"借腹生子"技术的发展,同时能加速鱼类优良珍稀物种的保存及增殖。本研究通过显微注射将锦鲤精原干细胞移植到斑马鱼幼体中,探索锦鲤精原干细胞在斑马鱼性腺中的发育过程:首先用酶消化方法从锦鲤幼体获得精原干细胞,进行荧光染色后,通过显微注射方法移植到刚孵化出膜斑马鱼受体的腹腔内;然后通过荧光标记追踪观察移植细胞在斑马鱼体受体性腺中的整合、增殖及分化情况。实验结果表明:通过酶消化法分离而得的性腺细胞主要为精原干细胞;荧光标记的锦鲤供体细胞移植入斑马鱼幼体腹腔后,在移植后第17天仍能检测到,并与斑马鱼内源生殖细胞共分布于幼鱼性原基。
Germ cells transplantation(GCT) is a technique whereby the donor germline stem cells are transplanted into the genital ridge, then differentiate into mature gametes. The application of GCT makesurrogatebroodstock production feasible and facilitate the preservations of the endangered species. In this study, the spermatogonial stem cells(SSCs) from Cyprinuscarpiowere transplanted into zebrafish host and traced during developing in the host: Firstly, SSCs of Cyprinuscarpio were dissociated by enzyme and labeled with fluorescence as donor cells. The cells were transplanted into the genital ridge of hatched zebrafish larvae. Then the development transplanted SSCs were examined, including the incorporation, proliferation and maturation in the host gonads. The results showed that the isolated cells majorly contained SSCs and the transplanted cells could be observed at 17 days post transplantation,co-localizing with the endogenous germ cells ofthe zebrafishhost.
