NMR技术检测高糖刺激对大鼠星形胶质细胞的代谢影响

摘要
在糖尿病患者体内,升高的血糖会导致中枢神经系统损伤,最终导致认知功能障碍~([1])。近年来,越来越多的研究开始关注星形胶质细胞在中枢神经系统中的重要作用~([2])。本工作通过高糖刺激原代培养的大鼠皮层星形胶质细胞,并运用核磁共振代谢组学方法检测其代谢变化。GFAP免疫荧光染色结果显示(图1),高糖刺激会导致胶质增生。时间轨迹图显示(图2),高糖组在24h至48h代谢轨迹明显不同。统计学结果显示(图3),高糖组Lac和ATP含量明显升高,说明能量代谢增强,且能量来源主要通过无氧酵解,而Asp含量显著性降低,进一步说明有氧氧化受到抑制。此外能量底物Ile,Val及Trp的消耗,起到补充TCA循环的作用。综上所述,星形胶质细胞在受到高糖刺激时,主要表现为能量代谢紊乱。
An elevated level of glucose has been found in diabetes patients associated with several central nervous system impairments.We attempted to explore the potential metabolic mechanism of HG-induced metabolic changes on primary cultured cortical astrocytes using NMR-based metabolomics.Here we show that the metabolic changes were mainly occurred between 24 h and 48h(Fig 2).Furthermore,the quantitative results may indicate the increase of anaerobic glycolysis and the reduction of TCA cycle(Fig 3).In conclusion,our results revealed that astrocytes may experience an energy metabolism disorder when exposed to high glucose.
