氟吗啉对大鼠内分泌干扰效应的初步研究

氟吗啉对SD大鼠的内分泌干扰效应的研究
    
    近年来,环境内分泌干扰物对人体健康和生态环境的影响受到了科学家和公众的广泛关注,农药也是其中的一类,而且与人类生活密切相关。氟吗啉是我国新开发的一种杀菌剂,它是否对哺乳类动物——SD大鼠具有内分泌干扰效应,是一个值得探讨的问题。因此我们开展了氟吗啉对SD大鼠的内分泌干扰效应的评价,为其合理使用提供科学依据。本研究以97.6 %氟吗啉原药为实验材料,分别设计15、45、和150 mg/kg/day三个给药剂量,以99.8%己烯雌酚作为阳性对照,其给药剂量为0.01 mg/kg/day。采用灌胃的染毒方式,进行了大鼠一代繁殖实验,通过一系列指标的检测来评价氟吗啉对SD大鼠内分泌干扰效应的大小。实验结果表明:阳性对照组——己烯雌酚(DES)组精子数比空白对照组少近50%、精子畸形率增加1.8倍,雌鼠的黄体数与对照组相比有显著差异。氟吗啉使45mg/kg/day和150 mg/kg/day剂量组的雄鼠、雌鼠发生肝脏毒性,引起肝细胞脂肪变性。氟吗啉使45 mg/kg/day和150 mg/kg/day剂量组的雄鼠精子数降低、畸形率增加,但是不能确定氟吗啉对雄鼠具有雌激素效应。氟吗啉对雌鼠的子宫湿重、上皮细胞没有雌激素效应,对生殖系统、生殖力没有影响。在本试验条件下,雌鼠在孕期和哺乳期接触氟吗啉对子鼠的生长发育没有影响。
In recent years, the effects of endocrine disrupting chemicals on human health and the environment have caused a great deal of concern among scientists and the public. Some pesticides that disrupt the endocrine system are commonly used in domestic situations. Flumorph is a new pesticide, and its effect on the mammalian (SD rats) endocrine system has not yet been determined. The object of this study was to evaluate endocrine disruption by flumorph(97.6 %) in SD rats, as a guide to safe and effective use of this pesticide. Flumorph was administered to rats at 15, 45, and 150 mg/kg/day, with a control group treated with 99.8% DES at 0.01 mg/kg/day, to investigate endocrine disruption in the reproductive system of parents and offspring. Liver cell swelling was found in male and female rats treated with flumorph at 45 and 150 mg/kg/day。Sperm counts of the DES group has decreased by around 50% and sperm morphological abnormalities has increased 1.8 times. In rats treated with 45 and 150 mg/kg/day, sperm counts were decreased and there was a greater incidence of sperm morphological abnormalities. Oral intake of flumorph did not induce any changes in uterine wet weight or size of uterine epithelial cells, and did not cause changes in the reproductive system or in reproductive ability, indicating a lack of estrogenic effects. Flumorph had no effect on gestation and on post-birth lactation.
