乌恰阿魏活性成分的研究

摘要
阿魏是伞形科阿魏属(Ferula L.)植物的总称,是极具药用价值的一类独特植物资源。阿魏(药材)为我国各族人民长期用来防病、治病的常用中药之一。阿魏作为药用已经有1300多年的历史,其间历代本草或药典都有记载。这一古老的药物,在我国至今不衰,说明它在常见病、多发病的防治上,有确切的疗效。由于服用简便,所以在民间的应用愈来愈广泛。
     中国药典(2000年版一部)规定:阿魏系伞形科植物新疆阿魏(Ferula sinkiangensis K. M. Shen)或阜康阿魏(Ferula fukanensis K. M. Shen)的树脂,具消积、散痞、虫积腹痛的治疗。除药典规定种外,尚有一些近缘种在民间长期以来作为该药材被加以应用。
     本文就是对阿魏的其中一种,乌恰阿魏化学成分提取分离和结构鉴定进行研究。在综合考察了多种提取方法、分离技术的基础上,根据实验室现有设备条件,采用有机溶剂法进行乌恰阿魏的提取。根据药物特性,通过理论分析和实验确定了分离、纯化乌恰阿魏时的最佳柱层析材料,最佳温度,最佳洗脱剂等工艺的优化,得到了如下结论:
     1:在溶剂提取乌恰阿魏有效成分的工艺中,通过单因素试验和多种比较统计方法,对影响提取因素进行了分析。结果表明,溶剂提取乌恰阿魏有效成分的最佳工艺参数为:分别用95%的乙醇和75%的乙醇提取后浓缩合并提取浸膏;液料比为4.5:1;提取温度在33℃;每次提取时间为5h;提取次数为5次.
     2:分离并鉴定了8个化合物,其中1个是新化合物, 2个是首次从该种得到;从乌恰阿魏的根中分离鉴定了1个黄铜类化合物和1个萜类化合物和1个羟基蒽醌类化合物。分离出的黄酮类化合物、羟基蒽醌类化合物和萜类化合物都是首次从该种得到。
     3:本文还对阿魏属植物的化学成分和药理活性研究的最新进展进行了综述,为合理开发利用本属药用植物资源和寻找先导化合物提供了一定的依据。
     4:对实验结果进行了讨论。
The genus Ferula, a class of unique plant resource,has been used as an important drug to cure and defend disease for a long time. It was recorded on many important codex of Chinese traditional medicine such as”XINXIUBENCAO”. The FERULA was recorded as medicine in Chinese codex in past dynasties over 1300 years.The age-old medicine was used until today,which indicates that curative effect of the medicine is definite on prevention and cure of familiar illness.Due to taking it easily,folk people take the medicine more and more widely.
     Ferula sinkiangensis K. M. Shen and Ferula fukanensis K. M. Shen were used as a governmental herb,which was confirmed by the pharmacopoeia of Peoples Republic of China(the first sect in the version of 2000).they were used to cure the disease of dyspepsia,blood tumidness and insect in vivo,et al.Including Ferula sinkiangensis K. M. Shen and ferula fukanensis K. M. Shen,some other species in the genus Ferula were used as nongovernmental herb to have the same factions.In order to research and develop the abundant resource of Ferula species in our country, the systemic studies on them were carried out.
     The paper is about one kind of Ferula,According to the analysis of varities methods about extraction and separation, organic solvents was used to extract Ferula. According to the herb’specility,analysis in theory and experiment to determine primary condition in separation and purification. technology .he main results are described as below:
     1: The single factor tests and orthogonal experiment design, analysisi of variance, multiple comparision were applied to analysis the main influence of factors in extracting yield of Ferula. The influence factors were:extraction Ferula by 95% and 75% C2H5OH; solid-liquid ratio is 1:4.5; distillation by 5 times and each time by 5h.
     2: 8 compounds were isolated and identified from it,according to the research plan of the faculty.The structures of the seven compounds were identified or elucidated on the basis of spectral data,physical constants and chemical evidences.
     3: The advances in research family the genus Ferula.
     4: The results were discussed.
