丹槐银屑浓缩丸对豚鼠银屑病样模型组织VEGF、VEGFR含量的影响

摘要
目的:通过丹槐银屑浓缩丸对豚鼠银屑病样模型耳部组织血管内皮生长因子( vascular endothelial growth factor , VEGF)及其受体( vascular endothelial growth factor receptor,VEGFR)水平影响的实验研究,探讨丹槐银屑浓缩丸治疗银屑病的作用机制。
     方法:将70只豚鼠随机分成7组,每组10只,即:空白组、模型组、西药对照组(氨甲喋呤组)、成药对照组(复方青黛胶囊组)、以及丹槐银屑浓缩丸低剂量组、中剂量组、高剂量组。用5%普萘洛尔乳剂外涂豚鼠耳背制备银屑病样病理改变模型。采用双抗体夹心酶联免疫法测定组织VEGF,VEGFR含量,观察其与皮损表现的关系。所得数据用SPSS13.0软件进行统计分析,计算均数、标准差,计量资料采用T检验,方差分析。
     结果:丹槐银屑浓缩丸高、中、低剂量组VEGF水平下降不明显( P>0.05),VEGFR水平出现不同程度下降,特别是中剂量组与其它组差异显著(P<0.01或P<0.05)。
     结论:丹槐银屑浓缩丸可使组织VEGFR水平显著降低,进而抑制血管内皮细胞增生,改善豚鼠银屑病样模型的皮损变化。
Objective: To investigate the effect of“Danhuai Yinxie Concentrated Pill”(DYCP) on the level of vascular endothelial growth factor(VEGF)and its receptor VEGFR in psoriasis-like guinea pigs' cutaneous lesions, so as to explore the mechanism of DYCP’s treatment for psoriasis.
     Methods: 70 guinea pigs were randomly divided into 7 groups,10 in each group , namely:blank group, model group,western medicine group ( control group),chinese patent medicine group(control group), DYCP high, medium and low dose group(treatment group).The guinea pigs were made into models of psoriasis by external application of 5%propranolol cream at skin of ears. The contents of VEGF and VEGFR in cutaneous lesions were detected by ELISA, then study the relation between the contents of above detection and the manifestation of cutaneous lesions. The data were analyzed with SPSS13.0 software to calculate the mean, standard deviation. Measurement data using T test, analysis of variance.
     Results: DYCP high, medium and low dose group showed a higher level,having no statistical difference with all the other groups(P>0.05). The level of VEGFR was declined to different degrees,especially for the medium dose group(P<0.01 or P<0.05 ).
     Conclusion:“Danhuai Yinxie Concentrated Pill”could significantly reduce the level of VEGFR in cutaneous lesions, and then inhibit the proliferation of endothelial cells and improve cutaneous lesions of psoriasis-like guinea pigs.
