石南藤化学成分的研究

摘要
石南藤(Piper wallichii(Miq.)Hand.-Mazz.),系胡椒科胡椒属植物,常绿攀援藤本,主产于四川、湖南、云南等地。传统中医常以其地上干燥部分入药,性微温,味辛苦。有祛风湿,通经络,强腰膝,止咳平喘,活血止痛的功效。主治风寒湿痹,腰膝酸痛,阳萎,咳嗽气喘,痛经,跌打肿痛等。现代药理研究发现石南藤中分离得到的活性成分具有明显抑制血小板活化因子诱导的血小板聚集作用。
     我们通过活性筛选,发现石南藤二氯甲烷部位提取物具有促进PC12细胞分化的作用,为此对该部位的化学成分进行了进一步地分离,得到了18个化合物,并根据理化性质及波谱分析鉴定了它们的结构,其中包括12个新木脂素:异山蒟素C(1)、isofutoquinol A(2)、(+)-玉兰脂B(3)、(+)-海风藤酮(4)、(+)-burchellin(5)、7S,8S,1′R-△~(8′)-3,4,5′-三甲氧基-1′,4′-二氢-4′-氧-7.O.2′.8.1′-新木脂素(6)、(+)-licarinA(7)、eupomatenoid-7(8)、南藤素(9)、山蒟素B(10)、山蒟素C(11)、山蒟素D(12);5个酰胺类生物:N-异丁基-反-2-反-4-奎二烯酰胺(13)、二氢毕拨明宁碱(14)、毕拨明宁碱(15)、风藤酰胺(16)、假荜拨酰胺A(17)和1个木脂素galgravin(18)。其中化合物1为新化合物,命名为异山蒟素C;化合物2、5、6、7、8、10、12、16、17为首次从石南藤中分离得到;化合物13、14、15、16、17具有促进PC12细胞分化的活性。
Piper wallichii(Miq.) Hand.-Mazz.,belonging to the Piperaceae family,is a perennial plant that is widely distributed in Sichuang,Hunan,and Yunnan Province in China.The aerial part of this plant is used as an effective folk medicine in China to treat beriberoid disease,alleviate pain,dredge meridian passage,and improve sexual function.Modern pharmacology research has elucidated the constituents of this plant have significant anti-platelet-activating factor(PAF) activity.
     We find that the CH_2Cl_2 extract of Piper wallichii(Miq.) Hand.-Mazz.has neurotrophic activity for PC12 cell.Phytochemical investigation led to the isolation of eighteen compounds.On the basis of their characteristics and spectra,the structures of these compounds were established,including twelve neolignans:isohancinone C(1), isofutoquinol A(2),(+)-denudatin B(3),(+)-kadsurenone(4),(+)-burchellin(5),7S, 8S,1'R-Δ~(8')-3,4,5'-trimethoxy-1',4'-dihydro-4'-oxo-7.O.2'.8.1'-neolignan(6)、(+)-licarinA(7)、eupomatenoid-7(8)、wallichinine(9)、hancinone B(10)、hancinone C(11)、hancinone D(12);five amides:N-isobutyl-deca-trans-2-trans-4-dienamide(13)、dihydropiperlonguminine(14)、piperlonguminine(15)、futoamide(16)、retrofractamide A(17);and one lignan:galgravin(18).Compound 1,named isohancinone C,is a new neolignan and never found before.Compound 2,5,6,7,8,10,12,16 and 17 were isolated from this plant for the first time.Compound 13,14,15,16 and 17 have been shown neurotrophic activity for PC12 cell.
