加味酸栆仁汤对模拟运动性失眠大鼠血浆单胺类神经递质的研究

摘要
1.研究目的
     通过对运动性失眠模型大鼠进行加味酸枣仁汤灌胃治疗,观察其血浆单胺类神经递质含量的变化,探讨运动性失眠的产生机理和加味酸枣仁汤的作用机制,为解决运动性失眠问题寻求更多的干预方法和提供科学依据。
     2.实验方法
     选取健康雌性Wister大鼠60只,随机分为6组,即对照组、模型组、自然康复组(简称自康组)、安定组、加味酸枣仁汤低剂量组(简称低剂组)、加味酸枣仁汤高剂量组(简称高剂组),每组10只。除对照组不做任何处理之外,其他各组以5周大强度负重游泳训练+腹腔注射PCPA建立模拟运动性失眠模型。建模后,模型组大鼠摘眼球取血,加肝素抗凝待测;自康组大鼠用生理盐水灌胃,低剂组和高剂组大鼠分别给予加味酸枣仁汤7.1g/kg和14.2g/kg灌胃;安定组大鼠给予安定0.2mg/Kg灌胃,一共5天。灌胃治疗后将各组大鼠摘眼球取血,通过高效液相色谱-荧光检测法测定血浆单胺类神经递质的含量。
     3.实验结果
     3.1模型组大鼠经5周大强度负重游泳训练+腹腔注射PCPA建模后,与对照组相比,血浆5-HT及5-HIAA含量明显降低(P<0.05);血浆NE和DA含量极明显升高(P<0.001)。
     3.2与模型组相比,经加味酸枣仁汤灌胃治疗后,大鼠血浆5-HT及其代谢产物5-HIAA含量明显上升(P<0.01),NE和DA含量显著降低(P<0.01)。
     3.3对照组大鼠每周平均体重和食量逐渐增长,其他各组大鼠建模后体重和食量有所下降,经加味酸枣仁汤治疗后增加。
     4.结论
     4.1模型组大鼠血浆5-HT及其代谢产物5-HIAA含量降低,NE和DA含量上升,睡眠减少,觉醒增加,表明5周大强度负重游泳训练+腹腔注射PCPA建立运动性失眠动物模型成功。
     4.2加味酸枣仁汤治疗后,运动性失眠大鼠血浆5-HT及5-HIAA含量上升,NE和DA含量降低,表明加味酸枣仁汤可能是通过调节单胺类神经递质含量的途径来治疗运动性失眠的。
     4.3加味酸枣仁汤治疗运动性失眠疗效显著,治疗效果优于西药安定片,而且一定范围内量效关系明显。
1. Research Objectives
     Through to gavage treatment of sports insomnia rat model with Jiawei Suanzaoren Decoction,observe changes in plasma monoamine neurotransmitter content and explore the mechanism ofexercise-induced insomnia and treatment of Jiawei Suanzaoren Decoction, and then look formore intervention methods to solve exercise-induced insomnia and provide a scientific basis.
     2.The experimental method
     60female healthy wistar rats were randomly divided into6groups, that is, the control group,the model group, the spontaneous recovery group (referred to as self-health group), the diazepamgroup, Jiawei Suanzaoren Decoction low-dose group (low dose group), Jiawei SuanzaorenDecoction high-dose group (high dose group), each group of10.In addition to the control groupreceived no treatment, other groups were established simulation exercise insomnia modelthrough to five weeks strength weight loading swimming training and intraperitoneal injection ofPCPA. After modeling, pick the eyeball from model rats to take blood, add heparinanticoagulation for the await measuring; the self-health group rats irrigation stomach with saline,the low-dose group and high dose group rats were given Jiawei Suanzaoren Decoction7.1g/kgand14.2g/kg irrigation stomach; the diazepam group rats were given diazepam0.2mg/Kgirrigation stomach, a total of5days.After consecutive treatment,each group rats will beeyeballed to take blood and then measure plasma monoamine neurotransmitter levels through thehigh performance liquid chromatography-fluorescence detection.
     3.The experimental results
     3.1After the model group rates modeling by five weeks strength weight loading swimmingtraining and intraperitoneal injection of PCPA, compared with the control group, plasma5-HTand5-HIAA content decreased significantly (P <0.05); plasma NE and DA content is extremelyincreased significantly (P <0.001).
     3.2Compared with model group, after Jiawei Suanzaoren Decoction irrigation treatment, the ratsplasma5-HT and its metabolites5-HIAA content increased obviously (P <0.01), NE and DAcontent significantly reduced (P <0.01)
     3.3The weight and appetite of control group rats gradually growth on average every week. Theweight and appetite of other groups has declined after modeling, and then increased after JiaweiSuanzaoren Decoction irrigation treatment.
     4. Conclusion
     4.1The model rats plasma5-HT and its metabolites5-HIAA content decreased, NE and DAcontent increased and reduce sleep, awakening increase, the results show that the five weeksstrength weight loading swimming training and intraperitoneal injection of PCPA build athletic insomnia animal model success.
     4.2After JiaweiSuan zao ren Decoction treatment, athletic insomnia rats plasma5-HT and5-HIAA content increased, NE and DA content decreased, show that the JiaweiSuan zao renDecoction may be through the regulation monoamine neural transmitter content approach totreatment of exercise-induced insomnia.4.3JiaweiSuan zao ren Decoction in the treatment of athletic insomnia curative effect is distinct,the treatment effect is better than that of western medicine diazepam, and the dose-effectrelationship is obvious in a certain range.
