颈椎病Ⅰ号胶囊兔模型神经根周围炎症因子抑制作用的实验研究

摘要
目的:研究颈椎病兔模型在药物的实验干预下神经根局部组织炎症因子的变化,初步探讨颈椎病Ⅰ号胶囊对兔模型神经根局部炎症因子的作用机理,进而研究颈椎病的形成机理,为颈椎病Ⅰ号胶囊治疗颈椎病提供确切的实验依据。
     方法:成年健康家兔46只,随机分配至正常对照组、假手术组及造模组,采用彭宝淦、余家阔等造模方法,切断棘上、棘间韧带并持续低头位的方法进行造模,假手术组只切断棘上、棘间韧带,不予低头位处理。术后2月造模成功后,将造模组随机分配至中药治疗组、模型对照组,中药治疗组予以药物进行干预,模型对照组、假手术组及正常对照组予以生理盐水对比,并采用放免疗法测定治疗后2周、4周神经根局部炎症因子前列腺素E_2(PGE_2)、6--前列腺素F_(1a)(6-K-PGF_(1a))、组织胺(HE)含量的变化。
     结果:
     1.颈椎病Ⅰ号胶囊与模型对照组相比,能有效地抑制兔模型神经根局部炎症因子,(P<0.01)。
     2.采用彭宝淦、余家阔等的造模方法,造模死亡率低(9.09%),检测效度高,经统计学分析,造模组与正常对照组、假手术组有显著差异(P<0.01)。
     结论:
     1.颈椎病Ⅰ号胶囊能有效抑制兔模型神经根局部炎症因子,作用确切,与模型对照组、假手术组及正常对照组相比,效果明显。
     2.炎症是导致盘源性颈腰痛的主因,神经根型颈椎病的形成与自身免疫性亦有关。
     3.彭宝淦、余家阔造模方法是符合颈椎病形成机理的较好的造模方法。
Objective:To study the nerve roots topical inflammatory factor change of cervical spondylosis rabbit model with drug intervention,the mechanism of cervical spondylosis NO.1 capsule for nerve roots topical inflammatory factor of rabbit model were explored to study the pathogenesis of cervical spondysis,which supply experimental basis for cervical spondylosis NO.1 capsule on treatment of cervical spondylosis.
     Methods:46 adult rabbits were divided into false operation group, model -making group and normal group,to establish the model by the methods of Peng bao gan and Yu jia kuo's which resect the cervical supraspinous and interspinous ligaments.2 months after the the models were established,The model-making group were divide into medicine group and normal model group,in which the medicine group were intervued by medicine and other groups were intervued by Sodium Chloride.Then the content of rabbits topical nerve roots inflammatory factors such as E_2(PGE_2)、6-K-PGF_(1a)、HE were examined and comparaed by the method of radioimmunity after treated 2w 4w and 6w.
     Result:
     1.The content of rabbits topical nerve roots inflammatory factors in cervical spondylosis NO.1 capsule group were significantly lower comparaed with normal model group(P<0.01)
     2.The model estebilsh methods of Peng bao gan and Yu jia kuo has lower mortality and high effective power of detection,and has significant difference in statistics comparaed with false operation group and normal group(P<0.01)
     Conclusion:
     1.Cervical sponddylosis NO.1 capsule can limit the nerve roots topical inflammatory factor of rabbit model effectively,compared with normal group,the effect is exactly obvious.
     2.Inflammation is the main reason of cervicodynia and lumbodynia caused by pathological changes of discus.and also has relationship with autoimmunity
     3.The model-making method of Peng bao ganYu jia kuo is a good method witch consistent with the mechanism of cervical spondylosis
