纤维素酶高产菌株选育与玉米秸秆生产酒精工艺研究

摘要
本论文对纤维素酶菌株的筛选及其发酵条件、性质和菌株鉴定进行了初步研究,并在其基础上对玉米秸秆同步糖化发酵生产酒精的工艺进行了探索性研究。
     在菌株筛选方面以保定郊区玉米地和秸秆垛下的土壤以及顺平食用菌生产基地的污染蘑菇袋为菌株筛选的分离源,筛选出分解纤维素酶活较高的LW1菌株。该菌株在秸秆:麦麸=2∶3,料水比为1∶2,培养基起始pH6.0的培养基中培养72小时后,在pH4.8,温度50℃的柠檬酸缓冲液中酶解1小时的滤纸酶活为240mg.G/h.g(干曲),而对照菌株康氏木霉为210mg.G/h.g(干曲)。该菌初步鉴定为康氏木霉。
     在玉米秸秆生产酒精的工艺研究方面确定了玉米秸秆的预处理条件和同时糖化发酵工艺条件。预处理条件的确定即用15%的氢氧化钠溶液浸泡秸秆24小时后,过滤并用沸水洗去碱液,在75℃下干燥至恒重;用正交实验的方法确定了同时糖化发酵的工艺条件,即发酵温度32℃,发酵时间3天,入池pH5.0和纤维素酶浓度20Iu/g原料,酒醅中的酒精浓度可达3%。
Food storage and process engineering Wang Qian
    This paper base on real of documents had preliminary studied cellulose decomposition fungus screening, optimum ferment condition, property and strain identify. And base on it, make pilot study on com produce alcohol process by SSF.
    On strain screening aspect, soil in corn ground, down corn pile in suburb of Boding and staining mushroom pocket in domestic fungus production base in ShunPing for extract source, and screen higher cellulose activity stain LW1. This strain after cultivate 72h in corn and Maxwell bran ratio is 2:3, water content ratio is 1:2, initial incubation basis pH6.0, and it is filter paper enzymatic activity run to 240mg.G/h.g(stem leaven), through enzymolysis filter paper Ih in pH4.8, temperature 50~?.?C citric acid buffer solution, but control strain T.Koningii is 210mg.G/h.(stem leaven).This strain preliminary identify is T.Koningii.
    In corn produce alcohol processing, establish corn pretreatment and SSF conditions. Pretreatment condition is 15% NaOH solution steep com 24h, filtrate and take boiling washed alkali liquor, and drying to constant weight in 75~(.)C;take perpendicularity experimental method established optimum SSF process condition, namely is ferment temperature 32~(.)C, ferment time 3 days, entrance pool pH5.0 and cellulase concentration 201U/g, alcohol concentration to 3%.
